摘要
以凝结芽孢杆菌(Bacillus coagulans)CNJD为研究对象,生物量为考察指标,采用单因素试验考察发酵时间、发酵温度、接种量、初始p H值、装液量和摇瓶转速等发酵条件对凝结芽孢杆菌CNJD菌体生长的影响,在此基础上,采用单因素及响应面法对其发酵培养基进行优化。结果表明,最优发酵条件为发酵时间16 h,发酵温度55℃,接种量2%,初始pH值5.0,摇床转速180 r/min,装液量50 mL/250 mL;最优发酵培养基组分为:蔗糖48.59 g/L,氯化钙1.70 g/L,硫酸锰0.33 g/L,酵母浸粉10.00 g/L,氯化钠1.50 g/L,胰蛋白胨10.00 g/L。在此优化工艺下,凝结芽孢杆菌CNJD的生物量达到7.86×10^(10)CFU/m L。
Taking Bacillus coagulans CNJD as the research object and biomass as the investigation index,the effects of fermentation conditions such as fermentation time,temperature,inoculum,initial pH value,medium volume and shaker speed on the growth of B.coagulans CNJD were explored by single factor experiments.On this basis,the fermentation medium was optimized by single factor and response surface methodology.The results showed that the optimal fermentation conditions were fermentation time 16 h,temperature 55℃,inoculum 2%,initial pH value 5.0,shaker speed 180 r/min,and medium volume 50 ml/250 ml.The optimal medium compositions were as follows:sucrose 48.59 g/L,calcium chloride 1.70 g/L,manganese sulfate 0.33 g/L,yeast extract 10.00 g/L,sodium chloride 1.50 g/L,and tryptone 10.00 g/L.Under the optimized process conditions,the biomass of B.coagulans CNJD reached 7.86×1010 CFU/ml.
