摘要
目的观察天香丹对急性心肌梗死大鼠血清中白细胞介素-18(IL-18)以及对梗死心肌细胞与心肌超微结构的影响,探索天香丹改善心肌梗死的作用机制。方法将54只7周龄左右雄性Wistar大鼠随机分为空白组、假手术组、模型组各6只,天香丹低剂量组、中剂量组、高剂量组各12只。造模成功后分别予低、中、高剂量的天香丹药物进行干预。检测大鼠血清中IL-18以及内皮素(ET)、一氧化氮(NO)的含量,电镜观测各组大鼠心肌细胞与心肌超微结构的变化,超声观测各组左室射血分数与心室结构改变。结果相对于模型组,天香丹高剂量组IL-18含量显著降低,NO含量显著升高(P<0.05)。镜下观察,与模型组比较,天香丹高剂量组可减轻炎细胞浸润、心肌细胞空泡变性及坏死程度,明显减少间质纤维组织增生。心脏超声检测天香丹高剂组对模型大鼠左室射血分数及心室结构有显著改善作用。结论天香丹是改善急性心肌梗死的有效药物,其作用机制可能与其抗炎,改善血管内皮功能及心功能与心肌超微结构有关。
Objective:To observe the effect of Tianxiangdan Pills on serum IL-18 and myocardial cell and myocardial ultrastructure in rats with acute myocardial infarction,and to explore the mechanism of Tianxiangdan Pills in improving myocardial infarction.Methods:Fifty-four 7-week-old male wistar rats were randomly divided into 6 groups:blank group,sham operation group,and model group with 6 rats each;model group plus drug inter⁃vention group(low dose group,middle dose group and high dose group)with 12 rats each.After successful model⁃ing,low,medium and high doses of Tianxiangdan Pills were given to intervene.The contents of IL-18,ET and NO in the serum of rats with acute myocardial infarction were detected.The ultrastructural changes of myocardial cells and myocardium were observed by electron microscope,and the left ventricular ejection fraction and ventricular structure were observed by ultrasound.Results:Compared with the model group,the content of IL-18 in the high dose group decreased significantly and the content of NO increased significantly(P<0.05).Microscopically,com⁃pared with the model group,the high dose group could reduce inflammatory cell infiltration,myocardial cell vacuo⁃lation and necrosis,and obviously reduce interstitial fibrous tissue proliferation.Cardiac ultrasound examination showed that the high dose group could significantly improve the left ventricular ejection fraction and ventricular structure of model rats.Conclusion:Tianxiangdan Pills is an effective drug to improve acute myocardial infarc⁃tion,and its mechanism may be related to its anti-inflammation,improvement of vascular endothelial function,car⁃diac function and myocardial ultrastructure.
