摘要
目的本研究旨在探析PGF2α和PGD2对体外绵羊卵巢颗粒黄体化细胞溶解及其相关基因差异表达的影响,为解析绵羊发情分子调控机制提供依据。方法体外培养哈萨克母羊卵巢颗粒细胞经黄体化处理后分别添加PGF2α、PGD2、PGD2+PGF2α进行诱导,采用ELISA技术检测P4浓度以及采用qRT-PCR技术检测黄体溶解相关基因mRNA的表达水平。结果结果显示,与对照组(颗粒黄体化细胞)相比,PGF2α组、PGD2组、PGD2+PGF2α组中P4浓度在24h显著降低,尤其是PGD2+PGF2α组呈现极显著下降变化(P<0.01);与对照组相比,PGF2α组、PGD2组中的StAR、3β-HSD呈现显著下降变化(P<0.05),PGD2+PGF2α组中的StAR、3β-HSD呈现极显著下降变化(P<0.01);PGF2α组中的PGFS无明显变化,PGD2组、PGD2+PGF2α组中的PGFS呈现极显著上升变化(P<0.01);PGD2组中的FP呈现显著上升变化(P<0.05),PGF2α组、PGD2+PGF2α组中的FP呈现极显著上升变化(P<0.01);PGD2+PGF2α组中的HPGDS呈现显著上升变化(P<0.05),PGF2α组、PGD2组中的HPGDS呈现极显著上升变化(P<0.01);PGF2α组、PGD2组、PGD2+PGF2α组中的DP呈现极显著上升变化(P<0.01)。结论PGF2α和PGD2均能够促进体外卵巢颗粒黄体化细胞的退化过程,但PGF2α和PGD2共同作用时,则在促溶黄体效应中呈现出显著的协同效应,这为全面认识母畜发情周期调节机理、优化母畜高效繁殖技术(尤其是PG方案)应用提供新的理论依据。
Objective This study aimed to explore the effects of PGF2αand PGD2 on the lysis of luteinized ovarian granulosa cells in sheep and the differential expression of related genes in vitro,and to provide a basis for analyzing the molecular regulation mechanism of sheep estrus.Methods In vitro cultured Kazakh ewe ovarian granulosa cells were treated with luteinization and induced by adding PGF2α,PGD2,PGD2+PGF2α.The P4 concentration was detected by ELISA and the mRNA expression level of luteinization-related genes was detected by qRT-PCR.Result The results showed that compared with the control group(granular luteinized cells),the P4 concentration in the PGF2αgroup,PGD2 group,and PGD2+PGF2αgroup decreased significantly at 24h,especially the PGD2+PGF2αgroup showed a very significant decrease(P<0.01);Compared with the control group,StAR and 3β-HSD in the PGF2αgroup and PGD2 group showed a significant decrease(P<0.05),and the StAR and 3β-HSD in the PGD2+PGF2αgroup showed a very significant decrease(P<0.01);PGFS in the PGF2αgroup did not change significantly,the PGFS in the PGD2 group and the PGD2+PGF2αgroup showed a very significant increase(P<0.01);the FP in the PGD2 group showed a significant increase(P<0.05),the PGF2αgroup,PGD2 The FP in the+PGF2αgroup showed a very significant increase(P<0.01);the HPGDS in the PGD2+PGF2αgroup showed a significant increase(P<0.05),and the HPGDS in the PGF2αgroup and the PGD2 group showed a very significant increase(P<0.01);the DP in the PGF2αgroup,PGD2 group,and PGD2+PGF2αgroup showed a very significant increase(P<0.01).Conclusion Both PGF2αand PGD2 can promote the degeneration process of ovarian granule luteinized cells in vitro,but when PGF2αand PGD2 work together,they exhibit a significant synergistic effect in promoting the luteinizing effect.This is a comprehensive understanding of the mechanism and optimization of the regulation of female animals’estrus cycle The application of high-efficiency breeding technology for female animals(especially the PG program)provides a new theoretical basis.
