氨氮急性胁迫下二、三倍体雌性虹鳟应激耐受性比较 被引量:2

Comparison of tolerance between diploid and triploid female rainbow trout under acute ammonia nitrogen stress

摘要 在集约化养殖条件下,当受到环境因子或人为因子胁迫时,三倍体雌性虹鳟死亡率明显高于二倍体雌性虹鳟,此现象与其应激耐受性有关。为了解氨氮急性胁迫下二倍体和三倍体雌性虹鳟应激耐受性差异,采用96 h半静态毒性试验法,在氨氮浓度分别为16.42、24.3、36.06 mg·L-1胁迫下,对二者急性毒性效应、血液生理生化指标以及肝脏非特异性免疫相关基因表达开展研究。结果表明,与二倍体雌性虹鳟相比,三倍体雌性虹鳟毒性反应较迅速且强烈,其血液中白细胞(WBC)、皮质醇(COR)和丙二醛(MDA)含量高;红细胞(RBC)、血红蛋白(HGB)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性低;鳃组织炎性细胞浸润,黏液细胞增多,鳃小片黏连,组织损伤严重;三倍体雌性虹鳟肝脏TNF-α、IL-1β、IL-6和IL-8基因表达量高于二倍体雌性虹鳟,HSP70、HSP90基因表达量低于二倍体雌性虹鳟。可知,三倍体雌性虹鳟对氨氮胁迫反应较二倍体雌性虹鳟更为敏感,其抗氧化及携氧调节能力较弱,即三倍体雌性虹鳟对氨氮急性胁迫应激耐受性更差。 The mortality rate of triploid female rainbow trout was significantly higher than that of diploid rainbow trout,when stressed by environmental or anthropogenic factors under intensive culture conditions,and this phenomenon was related to their stress tolerance.To understand the difference in stress tolerance between diploid and triploid female rainbow trout under acute stress of ammonia nitrogen,the acute toxicity effects,blood physiological and biochemical parameters and liver nonspecific immune-related gene expressions were investigated by 96 h semi-static toxicity assay at ammonia concentrations of 16.42,24.32 and 36.06 mg·L^(-1),respectively.The results showed that the acute toxic effects,blood physiological and biochemical indices and liver non-specific immune-related gene expression of the two species were similar to those of diploid female.The results showed that the toxic effects were more rapid and intense in triploid female rainbow trout compared to diploid females,with high levels of white blood cells(WBC),cortisol(COR)and malondialdehyde(MDA)in their blood.Red blood cells(RBC),hemoglobin(HGB),superoxide dismutase(SOD)and catalase(CAT)activities were low;gill tissue was infiltrated with inflammatory cells,increased mucus cells,gill lamellae adhesion and severe tissue damage;liver TNF-α,IL^(-1)β,IL-6 and IL-8 gene expression was high and HSP70 and HSP90 gene expression was low.The experimental results indicated that triploid female rainbow trout were more sensitive to ammonia and nitrogen stress than that of diploid,and their antioxidant and oxygen-carrying regulating abilities were weaker,which meant that triploid female rainbow trout was less stress tolerant to acute stress of ammonia and nitrogen.
