板栗根际高效解磷菌的筛选 被引量:11

摘要 【目的】从板栗根际分离筛选高效解磷微生物,研究解磷微生物对板栗生长的影响。【方法】选用磷酸钙作为培养基中唯一磷源,采用透明圈法结合钼蓝比色法筛选菌株,对所筛选菌株的解磷特性进行初步研究,并回接至盆栽板栗组培苗根际土壤,研究其促生作用。【结果】培养6 d后,共筛选得到具有解磷能力的菌株7株。对比各菌株的解磷能力,结果表明,菌株P6在以无机磷磷酸钙为磷源的培养基上溶磷指数为2.00,可溶磷含量达到75.98 mg/L,确定菌株P6为高效解磷菌。通过形态观察、生理生化特性检测及16S rDNA分子水平鉴定,确定菌株P6为洋葱伯克霍尔德氏菌Burkholderia cepacia。以有机磷卵磷脂为磷源对菌株P6进行液体培养,溶液中可溶磷含量为40.30 mg/L,碱性磷酸酶和酸性磷酸酶活性比CK分别增加了16.62、24.61 mmol/(L·h)。盆栽回接试验结果表明,接种菌株P6后板栗株高、茎粗分别比CK增加了60.70%、21.67%,植物地上部分和地下部分含磷量分别比CK提高了53.41%、52.33%,土壤碱性磷酸酶和酸性磷酸酶活性均显著增加,分别比CK增加了145.54%、449.27%,表明接种菌株P6对板栗解磷能力有明显的促进作用。【结论】菌株P6具有高效解磷能力,回接菌株P6能明显促进板栗组培苗的生长。 【Objective】In order to study the effect of phosphate solubilizing microorganisms on the growth of chestnut,microorganisms capable of efficient phosphate solubilizing were isolated and screened from the rhizosphere of chestnut.【Method】The calcium phosphate was selected as the sole phosphorus sources in the medium,and the phosphate solubilizing characteristics of the selected strains were preliminarily studied by transparent circle combined with molybdenum blue colorimetric method.And then inoculate to potted chestnut tissue rhizosphere soil,to study the growth-promoting effect.【Result】After culture of six days,seven strains with the ability of P-solubolizing were screened.Comparison of the P-solubolizing ability of the isolated strains,strain P6 showed high P-solubolizing ability with index 2.00,and the concentration of soluble phosphorus was 75.98 mg/L on the medium culture with inorganic phosphorous calcium phosphate as the sole phosphorus source.These results indicated that strain P6 was a high-efficiency P-solubolizing bacteria.According to the morphological observation,physiological and biochemical characteristics and 16S rDNA molecular identification,the strain P6 was identified as Burkholderia cepacia.The organophosphorus lecithin was used as the sole phosphorus source in liquid medium to culture the strain P6.The soluble phosphorus concentration in the medium increased to 40.30 mg/L and the activity of alkaline phosphatase and acid phosphatase were 16.62 and 24.61 mmol/(L·h)respectively more than CK.The pot experiment of inoculate strain P6 showed that the chestnut plant height and stem diameter increased 60.70%and 21.67%respectively,and the phosphorus concentration in the above-ground and underground parts of the plant increased 53.41%and 52.33%respectively,and the activities of alkaline phosphatase and acid phosphatase in soil also increased 145.54%and 449.27%respectively than CK.These indicated that return inoculating of strain P6 could obviously promote the phosphorus-releasing in chestnut.【Conclusion】Strain P6 has the ability of high-efficiency P-solubolizing and the return inoculation of strain P6 could obviously promote the growth of chestnut tissue culture seedlings.
