摘要
目的研究山楂叶总黄酮(HLF)对高脂血症(HLP)大鼠心脏的保护作用及其机制。方法取48只SD雄性大鼠随机分为空白组、模型组、辛伐他汀对照组(8mg/kg)、HLF低、中、高剂量组(100、200、400mg/kg),每组8只,除空白组外,其余各组采用高脂饲料喂养8周建立HLP大鼠模型。8周后,大鼠处死、取材,计算心脏指数,测定有关生化指标,采用HE病理切片染色观察心肌组织病理学变化,Western blot法测定心肌组织AMPK、p-AMPK、eNOS、p-eNOS蛋白表达水平。结果与空白组比较,模型组大鼠血浆CK、LDH、NEFA、MDA含量升高(P<0.01),而GSH、GSH-px、SOD含量降低(P<0.01),心肌AMPK、p-AMPK、eNOS、p-eNOS表达减少(P<0.01);与模型组比较,HLF高剂量组和辛伐他汀对照组CK、LDH、NEFA、MDA含量降低(P<0.05),GSH、GSH-px、SOD含量升高(P<0.01),给药各组AMPK、eNOS蛋白表达无明显差异,而p-AMPK、p-eNOS蛋白表达降低(P<0.05)。结论HLF可降低HLP大鼠血脂,其机制可能与HLF抗氧化作用以及调节心肌组织中p-AMPK、p-eNOS的蛋白表达升高有关。
Objective To study the protective effect and mechanism of Hawthorn Leaves Flavonoids(HLF)on the heart of hyperlipidemia rats.Methods Forty-eight SD male rats were randomly divided into blank group,model group,simvastatin control group(8 mg/kg),hawthorn leaves flavonoids low,medium and high dose groups(100,200,400mg/kg),with 8 rats in each group.The hyperlipidemia rat model was induced by high-fat diet except for the blank group.After 8 weeks,the rats were killed and the materials were collected.The cardiac related index was calculated,the biochemical indexes were determined,the pathological changes of heart tissue were observed by HE staining,and the expression levels of AMPK,p-AMPK,eNOS and p-eNOS protein were determined by Western blot.Results Compared with the blank group,the plasma levels of CK,LDH,NEFA and MDA in the model group were significantlty increased(P<0.01),while the contents of GSH,GSH-px and SOD were decreased(P<0.01).Compared with the blank group,the expression of AMPK,p-AMPK,eNOS and p-eNOS in the model group was decreased(P<0.01).Compared with the model group,the levels of CK,LDH,NEFA and MDA of the high-dose group of hawthorn leaf flavonoids and simvastatin control group were decreased(P<0.05),the contents of GSH,GSH-Px and SOD were increased(P<0.01).There was no significant difference in the expression of AMPK and eNOS protein in each group,and the expression of p-AMPK and p-eNOS protein was decreased(P<0.05).Conclusion Hawthorn leaves flavonoids can reduce blood lipids in hyperlipidemia rats,and its mechanism may be related to the antioxygenation and the increased protein expression of p-AMPK and p-eNOS in the heart.
