摘要
该文以过硫酸铵(ammonium persulfate,APS)、聚乙烯醇(polyvinyl alcohol,PVA)、3-氨基苯硼酸盐酸盐(3-aminophenylboronic acid hydrochloride,ABA)、苯胺(aniline,AN)为原料,合成聚苯胺基超分子水凝胶(polyaniline based supramolecular hydrogels,PPH)。以戊二醛(glutaraldehyde,GA)为交联剂制备聚苯胺基超分子水凝胶-戊二醛(polyaniline based supramolecular hydrogels-glutaraldehyde,PPH-GA)复合材料作为酶的固定化载体。利用3,5-二硝基水杨酸(dinitrosalicylic acid,DNS)法测定固定化果胶酶活力,计算固定化酶的载酶量,并用单因素变量法考察固定化酶和自由酶在不同酶浓度、温度、pH值、反应时间、果胶浓度下的相对活性,研究固定化果胶酶的酶催化性能。结果表明,固定化酶的最佳催化条件:pH 3.5、反应温度40℃、酶浓度3%、反应时间120 min、果胶浓度2%,此时载酶量为268.1 mg/g。
The supramolecular assembly of polyaniline hydrogel(PPH)was prepared using ammonium persulfate,polyvinyl alcohol,3-aminobenzeneboronic acid hydrochloride,and aniline.Polyaniline based supramolecular hydrogel-glutaraldehyde(PPH-GA)was synthesized using glutaraldehyde as a crosslinking agent.The activity of immobilized pectinase was determined by the dinitrosalicylic acid(DNS)method.The main parameters studied were the following:effects of pH,pectinase concentration,pectin concentration,reaction time,and effect of reaction temperature on the catalytic performance of the immobilized enzyme.The results showed that the immobilized enzyme was 268.1 mg/g at pH 3.5,reaction temperature 40℃,enzyme concentration 3%,reaction time 120 min,pectin concentration 2%.
