摘要
目的:探析慢性阻塞性肺疾病(COPD)大鼠血液、支气管灌洗液中水通道蛋白-1(AQP)-1及p-c-Jun-N-末端激酶(p-JNK)蛋白表达情况。方法:48只SPF级Wistar雄性大鼠随机平分为两组,模型组采用气管内注射脂多糖与被动吸烟法建立COPD大鼠模型,对照组在清洁环境下进行正常饲养,检测血液、支气管灌洗液中AQP-1、p-JNK表达情况并进行相关性分析。结果:模型组造模第4周、第8周、第12周的二氧化碳分压(PaCO_(2))高于对照组(均P<0.05),氧分压(PaO_(2))低于对照组(P<0.05),模型组造模第4周、第8周、第12周的第0.3秒用力呼气容积(FEV0.3)、第0.3秒用力呼气容积占用力肺活量的比值(FEV0.3/FVC)均低于对照组(均P<0.05)。模型组造模第4周、第8周、第12周的血液、支气管灌洗液AQP-1、p-JNK表达水平均高于对照组(均P<0.05)。在模型组中,Pearson相关分析造模第12周的血液、支气管灌洗液AQP-1、p-JNK表达水平与PaCO_(2)、PaO_(2)、FEV0.3、FEV0.3/FVC存在相关性(均P<0.05)。结论:COPD大鼠伴随有血液、支气管灌洗液的AQP-1、p-JNK高表达,且与大鼠的血气指标与肺功能存在相关性,从而参与COPD的发生与发展。
Objective:To investigate and analysis the expression of aquaporin-1(AQP-1)and p-c-Jun-N-terminal kinases(p-JNK)in blood and bronchial lavage fluid of rats with chronic obstructive pulmonary disease(COPD).Methods:48 SPF wistar male rats were equally randomly divided into control group and model group.The model group was adopted intratracheal injection of lipopolysaccharide and passive smoking to establish COPD rat model.The control group was fed normally in clean environment.The expression of AQP-1 and p-JNK in blood and bronchial lavage fluid was detected and the correlation analysis was conducted.Results:The partial pressure of carbon dioxide(PaCO_(2))at the 4th,8th,and 12th week of the model group were higher than those in the control group,and the partial pressure of oxygen(PaO_(2))were lower than those in the control group(all P<0.05).The FEV0.3 and FEV0.3/FVC of the model group at the 4th,8th,and 12th week were lower than those in the control group(all P<0.05).The expression levels of AQP-1 and p-JNK in the blood and bronchial lavage fluid of the model group were higher than those in the control group at the 4th,8th,and 12th week(all P<0.05).In the model group,Pearson correlation analysis showed that the blood and bronchial lavage fluid AQP-1 and p-JNK expression levels were correlated with PaCO_(2),PaO_(2),FEV0.3,FEV0.3/FVC at the 12th week of modeling(all P<0.05).Conclusion:COPD rats are accompanied by high expression of AQP-1 and p-JNK in blood and bronchial lavage fluid,which are related to the blood gas indicators and lung function of rats,and thus participate in the occurrence and development of COPD.
