青紫蓝兔遗传多态性统计分析

Statistical analysis of genetic polymorphism in chinchilla rabbits

摘要 目的统计分析青紫蓝兔的遗传多态性状况。方法应用微卫星DNA标记方法对27个位点在3组(A、B、C)共94个样品进行遗传多态性参数统计,主要包括等位基因数、观察杂合度(observed heterozygosity,Ho)、期望杂合度(expected heterozygosity,He)、多态信息含量(polymorphism information content,PIC)等,并应用SPSS 17.0软件分析各组遗传多态性参数统计学上的差异性,最后应用NTSYS软件构建3组非加权组平均法(unweighted pair-group method with arithmetic means,UPGMA)聚类图。结果94个样品在27个微卫星位点共发现74个等位基因,平均每个微卫星位点2.74个,Ho平均为0.80;He平均为0.55,PIC平均为0.47。A、B、C组彼此之间以及与总群体间的各项遗传多态性参数,组间差异均无统计学意义(P>0.05)。UPGMA聚类图结果显示,除C组89号外其余个体间相关系数均处于0.68~1.00。结论3组青紫蓝兔的遗传结构相似度极高,群体经过多代繁育后依然符合封闭群实验动物的要求,遗传质量控制比较稳定。因此,日常动物供应可以从任意一组中选取,不会影响生物制品热原检定实验结果的判定。 Objective To count and analyze the genetic polymorphism of the company’s chinchilla rabbit. Methods The genetic polymorphism parameters of 27 loci in 94 samples from A,B,C groups were analyzed by microsatellite DNA markers, including allele number, observed heterozygosity(Ho), expected heterozygosity(He) and polymorphism information content(PIC), then the statistical differences in the genetic parameters of each groups were counted by SPSS 17.0 software. Finally, the UPGMA cluster maps of three groups were constructed using NTSYS software. Results A total of 74 alleles were found at 27 microsatellite Loci in 94 samples, with an average of 2.74 at each microsatellite, 0.80 at Ho, 0.55 at He and 0.47 at PIC. There was no significant difference(P>0.05) among all genetic parameters among the three groups, and the three groups UPGMA cluster maps were successfully constructed with NTSYS,which showed that the correlation coefficients of all individuals were 0.68-1.00 except №.89 in C group. Conclusion The genetic structure of the A,B,C groups of chinchilla rabbits after breed for many generations are very similar, the population still meets the requirements of closed group experimental animals and the control of genetic quality is relatively stable, so the provided animals could be selected from any group, which does not affect the determination of pyrogen test results in biological products.
