酶解法提取山豆根多糖的工艺优化 被引量:2

摘要 为建立高得率的山豆根多糖提取工艺,试验以山豆根为原料,采用木瓜蛋白酶酶解法提取多糖。通过单因素试验和正交试验研究最佳提取工艺,采用苯酚-硫酸比色法测定多糖含量。结果表明:4个因素对山豆根多糖提取率的影响依次为酶浓度>酶解温度>酶解时间>酶解pH值。山豆根多糖最佳提取工艺条件为酶浓度2%、酶解温度55℃、酶解时间3 h、酶解pH值5,山豆根粗多糖含量为88.48%,多糖的提取率为5.11%,明显高于水提醇沉法提取的粗多糖得率(3.54%)。该方法简便、成本低、提取率高,适用于山豆根多糖的提取。 The experiment established a high-yield extraction process of Sophora subprostrate polysaccharide(SSP).In the experiment,the polysaccharide was extracted from Sophora subprostrate by papain.The optimum extraction process was studied by single factor test and orthogonal test.The content of polysaccharide was determined by phenol sulfuric acid colorimetry.The results showed that the effect of four factors on the extraction rate of SSP were enzyme concentration>enzyme temperatur>enzyme time>enzyme pH value.The optimum extraction conditions were enzyme concentration 2%,enzyme temperature 55℃,enzyme time 3 h and pH value 5.Under the optimum conditions,the content of polysaccharide was 88.48%,the extraction rate of SSP was 5.11%,which was significantly higher than that from water extraction and alcohol precipitation(3.54%).The method is simple,low cost and high extraction rate,which is suitable for extraction of SSP,and lays a foundation for the pharmacological study of SSP.
