摘要
目的分析程序性死亡受体-1(PD-1)和程序性死亡配体-1(PD-L1)在胃癌组织中的蛋白表达水平,探讨其与临床病理特征的相关性。方法选取2019年1月~12月甘肃省人民医院收治的46例胃癌患者(均实施胃癌根治术)为观察对象,收集其胃癌组织及癌旁组织,采用SP免疫组化和免疫印迹法检测胃癌组织及癌旁组织中PD-1和PD-L1的表达,并分析其临床意义。结果SP免疫组化检测显示,46例患者胃癌组织中PD-1蛋白表达阳性率为78.26%(36/46),PD-L1表达蛋白表达阳性率58.70%(27/46),差异有统计学意义(P<0.05)。不同TNM分期PD-1表达比较,差异有统计学意义(P<0.05);不同年龄、性别、肿瘤直径大小、分化程度、浸润分布情况、淋巴结转移PD-1表达比较,差异无统计学意义(P>0.05)。不同肿瘤浸润情况PD-L1表达比较,差异有统计学意义(P<0.05);不同年龄、性别、肿瘤大小、分化程度、TNM分期、淋巴结转移间PD-L1表达比较,差异无统计学意义(P>0.05)。WB检测显示,在胃癌细胞中PD-1和PD-L1均有表达。结论PD-L1和PD-1可在胃癌组织中高度表达,可将其作为诊断病情的有效指标。
Objective To analyze the protein expression levels of programmed death receptor-1(PD-1)and programmed death ligand-1(PD-L1)in gastric cancer tissues,and explore their correlation with clinicopathological characteristics.Methods 46 cases of gastric cancer patients(all underwent radical gastric cancer surgery)admitted to Gansu Provincial People’s Hospital from January to December 2019 were selected as observation objects,and their gastric cancer tissues and adjacent tissues were collected.SP immunohistochemistry and immunoblotting were used to detect the expression of PD-1 and PD-L1 in gastric cancer tissues and adjacent tissues,and analyze their clinical significance.Results SP immunohistochemical examination showed that the positive rate of PD-1 protein expression in 46 patients with gastric cancer tissue was 78.26%(36/46),and the positive rate of PD-L1 expression protein expression was 58.70%(27/46),the difference was statistically significant(P<0.05).Comparison of PD-1 expression in different TNM stages,the difference was statistically significant(P<0.05);There were no statistically significant differences in PD-1 expression among different ages,genders,tumor diameters,degrees of differentiation,infiltration distribution,and lymph node metastasis(P>0.05).Comparison of PD-L1 expression in different tumor infiltration conditions showed statistically significant differences(P<0.05);There was no statistically significant difference in PD-L1 expression among different ages,genders,tumor size,degree of differentiation,TNM staging,and lymph node metastasis(P>0.05).WB detection showed that both PD-1 and PD-L1 were expressed in gastric cancer cells.Conclusion PD-L1 and PD-1 can be highly expressed in gastric cancer tissues,and they can be used as effective indicators for the diagnosis of the disease.
