外周血miRNA-26b和miRNA-216a表达与支气管哮喘急性发作儿童肺功能的相关性 被引量:3

Correlation between the expression of miRNA-26b and miRNA-216a in peripheral blood and lung function in children with acute attack of bronchial asthma

摘要 目的探讨外周血微小核糖核酸(miRNA)-26b、miRNA-216a的表达和血清炎症因子水平与支气管哮喘急性发作患儿肺功能的相关性。方法选择广东省妇幼保健院2017年12月至2019年9月收治的支气管哮喘急性发作患儿107例作为急性发作组、支气管哮喘缓解期患儿93例作为缓解组,另选同期健康体检儿童90例作为对照组。采用实时荧光定量反转录聚合酶链反应(RT-qPCR)测定三组儿童外周血miRNA-26b和miRNA-216a表达;采用酶联免疫吸附法测定血清炎症因子[超敏C反应蛋白(hs-CRP)、白细胞介素(IL)-6和肿瘤坏死因子(TNF)-α]水平;采用肺功能仪测定第一秒用力呼气容积(FEV1)、最大呼气峰值流速(PEF)和用力肺活量(FVC)。比较各组外周血miRNA-26b和miRNA-216a表达、炎症因子水平和肺功能变化;分析miRNA-26b、miRNA-216a、炎症因子与肺功能的相关性。结果外周血miRNA-26b和miRNA-216a表达均呈急性发作组>稳定期组>对照组(P<0.05);炎症因子hs-CRP、IL-6和TNF-α水平均呈急性发作组>稳定期组>对照组(P<0.05);肺功能FEV1、PEF和FVC均呈急性发作组<稳定期组<对照组(P<0.05)。miRNA-26b、miRNA-216a、hs-CRP、IL-6、TNF-α与FEV1、PEF和FVC分别呈线性负相关(P<0.05)。结论支气管哮喘急性发作儿童外周血miRNA-26b和miRNA-216a高表达;miRNA-26b、miRNA-216a和炎症因子与肺功能水平呈线性负相关。 Objective To investigate the relationship between the expression of miRNA-26 b and miRNA-216 a in peripheral blood,the level of inflammatory factors in serum and lung function in children with acute attack of bronchial asthma.Methods From December 2017 to September 2019,107 children with acute attack of bronchial asthma were selected as the acute attack group,93 children with remission of bronchial asthma were selected as the remission group,and 90 healthy children were selected as the control group.The RT-qPCR was used to detect the expression of miRNA-26 b and miRNA-216 a in peripheral blood of three groups of children.ELISA was used to detect the level of inflammatory factors(hs-CRP,IL-6 and TNF-α)in serum.Pulmonary function analyzer was used to detect the forced expiratory volume in the first second(FEV1),peak expiratory flow(PEF)and forced vital capacity(FVC).The expressions of miRNA-26 b and miRNA-216 a in peripheral blood,the levels of inflammatory factors in serum and lung function were compared.The correlation between miRNA-26 b,miRNA-216 a,inflammatory factors and lung function was analyzed.Results The expression of miRNA-26 b and miRNA-216 a in peripheral blood increased from control group to stable group to acute attack group(P<0.05).The level of inflammatory factors(hs-CRP,IL-6 and TNF-α)increased from control group to stable group to acute attack group(P<0.05).The lung functions(FEV1,PEF and FVC)decreased from control group to stable group to acute attack group(P<0.05).miRNA-216 a、hs-CRP、IL-6、TNF-αwere negatively correlated with FEV1,PEF and FVC respectively(P<0.05).Conclusion The expression of miRNA-26 b and miRNA-216 a in peripheral blood of children with acute attack of bronchial asthma may be high.miRNA-26 b and miRNA-216 a and inflammatory factors are negatively correlated with lung function.
