摘要
阿尔兹海默症(AD)已成为严重危害人类健康的重大神经退行性疾病,因其发病机制尚未完全阐明,目前仍没有良好的临床治疗药物。随着系统生物学的发展,更多的发病机制不断被提出,为抗AD药物的研发提供了理论支持。本文作者对胆碱能损伤学说、Aβ学说、tau蛋白学说、炎症学说、兴奋性神经损伤学说、“微生物-脑-肠轴”学说等发病机制及临床相关药物的研究进展进行综述,旨在为抗AD药物的研发提供参考。
Alzheimer's disease(AD)has become a major neurodegenerative disease that seriously endangers human health.Because its pathogenesis has not been fully elucidated,there is yet no ideal clinical drug for effective treatment.With the development of systematic biology,more and more pathogeneses have been proposed,which offer the theoretical supports for the research and development of anti-AD drugs.This paper reviews the research progress of popular pathogenesis,such as cholinergic injury theory,Aβtheory,tau protein theory,inflammation theory,excitatory nerve injury theory,“microorganism-brain-gut axis”theory,and related clinical drugs as well,in order to provide a reference for the research of anti-AD drugs.
