摘要
目的探讨左旋含羞草碱对结直肠癌细胞增殖、凋亡的影响及分子机制。方法结直肠癌细胞SW480分为对照组、左旋含羞草碱低、中、高浓度组、pcDNA组、pcDNA-LATS1组、左旋含羞草碱+si-NC组、左旋含羞草碱+si-LATS1组。四甲基偶氮唑盐比色法(MTT)检测细胞增殖抑制率;蛋白质印迹(Western blot)法检测细胞周期蛋白D1(Cyclin D1)、细胞周期蛋白依赖性激酶抑制剂1A(p21)、B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X(Bax)、大肿瘤抑制基因1(LATS1)蛋白表达;流式细胞术检测细胞凋亡;实时荧光定量PCR(RT-qPCR)检测LATS1 mRNA表达水平。结果左旋含羞草碱低、中、高浓度处理的结直肠癌细胞SW480细胞增殖抑制率升高,细胞凋亡率升高,LATS1表达水平升高,且呈浓度依赖性(P<0.05)。过表达LATS1抑制结直肠癌细胞增殖,并促进细胞凋亡。抑制LATS1表达逆转了左旋含羞草碱对结直肠癌SW480细胞增殖和凋亡的作用。结论左旋含羞草碱可能通过上调LATS1表达抑制结直肠癌细胞增殖,并促进细胞凋亡。
Objective To explore the effect and molecular mechanism of L-mimosine on proliferation and apoptosis of colorectal cancer cells.Methods Colorectal cancer cells SW480 were divided into control group,low,medium,and high-concentration group of L-mitosine,pcDNA group,pcDNA-LATS1 group,L-mitosine+si-NC group,L-mimosine+si-LATS1 group.Methyl thiazolyl tetrazolium assay(MTT)was used to detect the cell proliferation inhibition rate.Western blot method was used to detect Cyclin D1,cyclin-dependent kinase inhibitor 1A(p21),B cell lymphoma/leukemia-2(Bcl-2),Bcl-2 related X(Bax),large tumor suppressor gene 1(LATS1)protein expression;flow cytometry to detect apoptosis.The real-time fluorescence quantitative PCR(RT-qPCR)was used to detect LATS1 mRNA expression.Results In colorectal cancer cells SW480 treated with low,medium and high concentrations of L-mimosaine,the cell proliferation inhibition rate,the apoptosis rate and the expression of LATS1 were increased(P<0.05).Overexpression of LATS1 inhibits the proliferation of colorectal cancer cells and promotes apoptosis.Inhibition of LATS1 expression reversed the effect of L-mimosine on proliferation and apoptosis of colorectal cancer SW480 cells.Conclusion L-Mimosaine may inhibit the proliferation of colorectal cancer cells and promote apoptosis by up-regulating the expression of LATS1.
