竹叶提取物对哺乳仔猪回肠形态和抗氧化功能的影响 被引量:6

摘要 [目的]本试验旨在研究竹叶提取物(BLE)对哺乳仔猪回肠肠道形态和抗氧化功能的影响。[方法]选取3日龄体重相近哺乳仔猪64头,随机分为4组,每组8个重复,每个重复2头哺乳仔猪。对照组每日灌喂羧甲基纤维素钠溶液,试验组分别灌喂100、200和300 mg·kg^(-1)的BLE-羧甲基纤维素钠溶液。试验期21 d。[结果]与对照组相比,灌喂300 mg·kg^(-1) BLE显著提高了回肠绒毛高度(P<0.05),但对隐窝深度无显著影响(P>0.05);灌喂200和300 mg·kg^(-1) BLE均显著降低仔猪血清和回肠黏膜丙二醛含量(P<0.05);灌喂300 mg·kg^(-1) BLE显著提高血清和回肠黏膜谷胱甘肽过氧化物酶(GSH-Px)的活性,并上调回肠黏膜核转录因子E2相关因子2(Nrf_(2))和GSH-Px mRNA的表达(P<0.05);灌喂300 mg·kg^(-1) BLE还可显著提高回肠黏膜超氧化物歧化酶(SOD)的活性,并上调SOD_(2)和血红素氧合酶1 mRNA的表达水平(P<0.05);此外,灌喂200和300 mg·kg^(-1) BLE均显著上调回肠黏膜Nrf_(2)和SOD_(2)蛋白的表达量(P<0.05)。[结论]竹叶提取物可以改善哺乳仔猪回肠肠道形态结构,促进哺乳仔猪肠道发育,并通过激活Nrf_(2)通路提高回肠黏膜抗氧化功能,且灌喂剂量为300 mg·kg^(-1)时具有较好的作用效果。 [Objectives]The aim of this study was to investigate the effects of bamboo leaf extract(BLE)on the morphological structure and antioxidant function of ileum in suckling piglets.[Methods]At 3 days of age,sixty-four piglets[Duroc×(Landrace×Yorkshire)]with similar body weight(BW)were randomly selected and divided into 4 groups with 8 replicates,and each replicate had 2 piglets.In the control(CON)group,piglets were treated orally with sodium carboxymethyl cellulose solution daily.Piglets in the BLE-treated groups received BLE by oral route daily at dosage of 100,200,300 mg·kg^(-1),respectively.The trial lasted 21 days.[Results]Compared with CON group,oral administration of BLE at dose of 300 mg·kg^(-1) significantly increased the villi height of the ileum(P<0.05),but had no significant effect on the crypt depth(P>0.05).Oral administration of BLE at doses of 200 and 300 mg·kg^(-1) significantly reduced the content of malondialdehyde in serum and ileum mucosa of piglets(P<0.05).Oral administration of BLE at doses of 300 mg·kg^(-1) significantly improved the serum and ileum mucosa glutathione peroxidase(GSH-Px)activities,and increased the mRNA expression level of nuclear factor erythroid 2-related factor 2(Nrf_(2))and GSH-Px in the ileum mucosa(P<0.05).Oral administration of BLE at dose of 300 mg·kg^(-1) could significantly improve the activity of superoxide dismutase(SOD)in the ileum mucosa,and increased the mRNA expression level of SOD_(2) and heme oxygenase 1(P<0.05).In addition,200 and 300 mg·kg^(-1) BLE increased the protein expression levels of Nrf_(2) and SOD_(2) in ileum mucosa(P<0.05).[Conclusions]BLE could improve the intestinal morphology and structure of suckling piglets,promote the intestinal development of suckling piglets,and improve ileum mucosa antioxidant capacity by activating Nrf_(2) pathway.The effects were better when the dose was 300 mg·kg^(-1).
