摘要
目的:探究桂枝茯苓丸对子宫腺肌病小鼠子宫组织形态及Rho A/ROCK信号通路相关分子的影响,明确桂枝茯苓丸治疗子宫腺肌病的药效物质基础。方法:雌性新生ICR小鼠90只,随机分组后,模型组及桂枝茯苓丸低、中、高剂量组于出生后第25天,连续每日滴喂他莫昔芬造模,饲养至49日龄时,每组随机抽取2只小鼠验证造模是否成功,验证后分组灌胃处理1个月后处死小鼠取子宫组织,行苏木素-伊红染色检测子宫病理变化并进行分级,荧光定量PCR检测Rho A mRNA、ROCK1 mRNA、ROCK2 mRNA表达水平,蛋白印迹检测p-MLC蛋白表达水平。结果:桂枝茯苓丸高、中剂量组小鼠子宫内膜浸润程度分级明显高于空白对照组(P<0.05);桂枝茯苓丸低剂量组小鼠子宫内膜浸润程度分级与空白对照组比较,差异无统计学意义(P>0.05);桂枝茯苓丸高、低剂量组小鼠子宫组织中Rho A mRNA、ROCK1 mRNA相对表达量,以及p-MLC蛋白表达明显低于模型组(P<0.05或P<0.01);桂枝茯苓丸高剂量组小鼠子宫组织中Rho A mRNA、ROCK2 mRNA相对表达量明显低于桂枝茯苓丸低剂量组(P<0.01)。结论:适宜剂量的桂枝茯苓丸可能通过调节子宫腺肌病小鼠Rho A/ROCK信号通路相关分子的表达,达到治疗ICR小鼠子宫腺肌病的目的。
Objective:To explore the effects of Guizhi Fuling Pills on uterine tissue morphology and Rho A/ROCK signaling pathway related molecules in mice with adenomyosis,and to clarify the effective material basis of Guizhi Fuling Pills in the treatment of adenomyosis.Methods:A total of 90 female newborn ICR mice were randomly divided into groups.The model group and the Guizhi Fuling Pills low,medium and high-dose groups were given continuous drips of tamoxifen every day for modeling and rearing on the 2 ndto 5 thday after birth.At the age of 49 days,2 mice were randomly selected from each group to verify whether the model was successful.After verification,the mice were sacrificed by intragastric administration for 1 month and the uterine tissues were collected.The uterine tissues were taken and the pathological changes of uterus were detected by hematoxylin eosin staining.The expression levels of Rho A mRNA,ROCK1 mRNA and ROCK2 mRNA were detected by fluorescence quantitative PCR,and the protein expression level of P-MLC was detected by Western blotting.Results:The grade of endometrial infiltration in Guizhi Fuling Pills high and medium-dose group was significantly higher than that in blank control group(P<0.05).There was no significant difference in endometrial infiltration grade between Guizhi Fuling Pills low-dose group and blank control group(P>0.05).The relative expressions of RhoA mRNA,Rock1 mRNA and P-MLC protein in uterine tissue of mice in Guizhi Fuling Pills high and low dose group were significantly lower than those in model group(P<0.05 or P<0.01).The relative expression of RhoA mRNA and Rock2 mRNA in uterine tissue of mice in Guizhi Fuling Pills high dose group was significantly lower than that in Guizhi Fuling Pills low dose group(P<0.01).Conclusion:The appropriate dose of Guizhi Fuling Pills may regulate the expression of RhoA/ROCK signaling pathway related molecules in adenomyosis mice to achieve the purpose of treating adenomyosis in ICR mice.
