摘要
目的茵陈在临床上用于肝炎的治疗具有疗效明确、不良反应少的特点。通过网络药理学的方法,筛选茵陈的主要活性成分,构建药物-成分-疾病-靶点网络,探讨其对肝炎作用的分子机制。方法采用中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)、Genecard、在线《人类孟德尔遗传》(Online Mendelian Inheritance in Man,OMIM)和String等数据库,获取茵陈所含的化学成分及靶点、肝炎疾病靶点和蛋白互作网络(protein-protein interaction,PPI)信息。通过Cytoscape 3.7.2构建药物-成分-疾病-靶点网络信息。基于生物信息学开源软件Bioconductor和R语言对茵陈作用的生物学过程和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)进行富集分析。结果茵陈的13个有效成分通过调控156个靶点和160条通路对肝炎进行治疗,且AKT1、IL6、VEGFA、JUN等靶蛋白是其治疗肝炎的关键基因。结论该研究基于网络药理学方法对茵陈治疗肝炎作用机制有了新的认识,阐明茵陈活性成分与肝炎的关系,为临床药物治疗提供了科学依据。
Objective Yinchen(Artemisiae Scopariae Herba)was used in anti-hepatitis,which has the characteristic of curative effect and low side effects.To predict the molecular mechanism of Yinchen(Artemisiae Scopariae Herba)in anti-hepatitis,the ingredients of it were filtered to establish the drugcomponent-disease-target network by the method of network pharmacology.Methods The components of Yinchen(Artemisiae Scopariae Herba),disease targets of hepatitis and protein-protein interaction networks were obtained via TCMSP,Genecard,OMIM and String database.The network information of the“medicine-component-disease-target”were established via Cytoscape 3.7.2 version.The gene ontology biology process and Kyoto Encyclopedia of Genes and Genomes pathway analysis were performed via Bioconductor and R-project.Results 13 active components of Yinchen(Artemisiae Scopariae Herba)could treat hepatitis by regulating 156 targets and 160 pathways,AKT1,IL6,VEGFA,JUN and other target proteins are the key genes in the treatment of hepatitis.Conclusion This research has a new knowledge of treating hepatitis via Yinchen(Artemisiae Scopariae Herba)by the method of network pharmacology,it has the ability to illuminate the relationship between hepatitis and Yinchen(Artemisiae Scopariae Herba),maybe could provide scientific evidence for clinical drug therapy.
