培元抗癌汤联合替加氟对肝癌肺转移肿瘤微环境中MMP2、MMP9及CXCL1、CXCL2调节的实验研究 被引量:7

摘要 目的观察培元抗癌汤联合替加氟对MMP2、MMP9及CXCL1、CXCL2调节作用及对H22肝癌细胞转移能力影响。方法用H22肿瘤细胞悬液,注入小鼠尾静脉中造模后进行药物干预。解剖小鼠,取出肺组织,镜下观察肺部转移结节数目并记录,计算肺转移抑制率。免疫组化法检测转移瘤中MMP2、MMP9蛋白表达水平,ELISA检测小鼠血中CXCL1、CXCL2的水平。结果空白组与荷瘤组、替加氟组、中西医组结节数比较有统计学意义(P<0.05),替加氟组与中西医组比较有统计学意义(P<0.05)。西药组、中西医组转移抑制率分别为11%、49%,中西医组疗效优于单药组。MMP2、MMP9在空白组呈弱阳性,在荷瘤组呈强阳性,在替加氟组、中西医组呈阳性,且中西医组表达更少。中西医组对肺转移癌外周血中CXCL1有抑制作用,替加氟组无抑制作用。西药组、中西医组均对肺转移癌外周血中CXCL2有抑制作用,两组数据均表明中西医组疗效优于单药组。结论培元抗癌汤联合替加氟在体内能抑制H22细胞肝癌细胞转移能力,其机制可能是调节肿瘤微环境中的MMP2、MMP9及CXCL1、CXCL2。 Objective To observe the effect of Peiyuan Kang'ai Decoction(培元抗癌汤)combined with tegafur on the regulation of MMP2,MMP9,CXCL1 and CXCL 2 and the influence on the metastasis of H22 cells.Methods H22 tumor cell suspension was injected into the tail vein of mice to establish the model.And then the lung tissue was taken out to observe the number of pulmonary metastasis nodules under the microscope.We recorded and calculated the inhibition rate of pulmonary metastasis.The expression levels of MMP2 and MMP9 were detected by immunohistochemistry and CXCL1 and CXCL2 were detected by ELISA.Results The number of nodules in the blank group was significantly higher than that in the tumor-bearing group,tegafur group and traditional Chinese and Western medicine group(P<0.05).The inhibition rate of metastasis in Western medicine group and traditional Chinese and Western medicine group were 11%and 49%,respectively.The curative effect of Western medicine group was better than that of single medicine group.MMP2 and MMP9 were weakly positive in blank group,strongly positive in tumor-bearing group,and less positive in tegafur group and traditional Chinese and Western medicine group.The CXCL1 in the peripheral blood of lung metastasis was inhibited in the group of traditional Chinese and Western medicine,but not in the group of tegafur.Western medicine group and Chinese and Western medicine group had inhibitory effect on CXCL2 in peripheral blood of lung metastasis cancer.The data ofgroups showed that the curative effect of Chinese and Western medicine group was better than that of single medicine group.Conclusion Peiyuan Kang'ai Decoction combined with tegafur can inhibit the metastasis of H22 cells in vivo.The mechanism may be to regulate MMP2,MMP9,CXCL1 and cxcl2 in tumor microenvironment.
