摘要
目的观察蒙医温针疗法对抑郁大鼠脑组织及血清中细胞因子IL-1β、IL-2、IL-6及TNF-α的影响。方法SD雄性大鼠60只,随机分为空白组、模型组、温针组、药物组,抑郁大鼠采用对慢性不可预知性应急刺激致抑郁大鼠模型,温针组采用蒙医温针疗法治疗,取顶会穴、赫依穴、心穴施针,药物组采用氟西汀治疗。取脑组织及血清测定IL-1β、IL-2、IL-6及TNF-α的含量。结果IL-1含量,蒙医温针能显著降低下丘脑、大脑前额皮层异常增加的IL-1含量,与模型组比较有显著差异(P<0.05,P<0.01);对海马IL-1含量略有降低作用,与模型组无明显差异(P>0.05)。IL-2含量,蒙医温针能显著降低下丘脑、海马、大脑前额皮层异常增加的IL-2含量,与模型组比较有显著性差异(P<0.05,P<0.05,P<0.01)。IL-6含量,蒙医温针能显著降低下丘脑、海马、大脑皮层异常增加的IL-6含量,与模型组比较有显著性差异(P<0.05,P<0.01,P<0.01);TNF-α含量,蒙医温针能显著降低下丘脑异常增加的TNF-α含量,与模型组比较有显著性差异(P<0.05);对海马、大脑皮层TNF-α含量无影响,与模型组比较无明显差异(P>0.05,P>0.05,P>0.05)。蒙医温针能显著降低血清中异常增加的IL-1、IL-2、IL-6的含量,与模型组比较有显著性差异(P<0.05,P<0.05,P<0.05)。结论蒙医温针疗法可调节抑郁大鼠脑组织和血清内与抑郁相关细胞因子含量,揭示了蒙医温针疗法改善抑郁的机制与调节免疫功能、调节具有抗抑郁作用的细胞因子含量有关。
Objective To observe the effects of Mongolian medical warm acupuncture on the levels of IL-1β,IL-2,IL-6 and TNF-αin brain tissue and serum of depression rats.Methods Sixty male SD rats were randomly divided into blank group,model group,warm acupuncture group and drug group.The rats were treated with CUMS stimulusto set up the depressionrats model.In the warm acupuncture group,Mongolian medical warmacupuncture therapy was used,and the acupuncture was applied at the Dinghui point,Heyipooint and Heart point.The drug group was treated with Fluoxetine Injection.Brain tissue and serum were taken to determine the contents of IL-1β,IL-2,IL-6 and TNF-α.Results The content of IL-1βwas significantly decreased by Mongolian medical warm acupuncturein the hypothalamus and prefrontal cortex,which was significantly different from that of the model group(P<0.05,P<0.01).The content of IL-1βin the hippocampus decreased slightly,but there was no significant difference from that of the model group(P>0.05).The content of IL-2 with abnormal increase in the hypothalamus,hippocampus and prefrontal cortexcan be significantly reduced by Mongolian medical warm acupuncture,which was significantly different from that of the model group(P<0.05,P<0.05,P<0.01).The content of IL-6 with abnormal increase in the hypothalamus,hippocampus and cerebral cortexcan be significantly reduced by Mongolian medical warm acupuncture,which was significantly different from that of the model group(P<0.05,P<0.01,P<0.01).Mongolian medical warm acupuncture can significantly reduce the content of TNF-αwith abnormal increase in hypothalamus,which was significantly different from that of the model group(P<0.05).There was no effect on the content of TNF-αin the hippocampus and cerebral cortex,and no significant difference from that of the model group(P>0.05,P>0.05,P>0.05).Conclusion Mongolian medical warm acupuncture can regulate the contents of deperession-related cytokines in brain tissue and serum of rats with depression,revealing that the mechanism of Mongolian medical warm acupuncture to improve depression is related to the regulation of immune function and the regulation of cytokines that have the function of anti-depression.
