摘要
2020年11月贵州毕节某猪场一头经产母猪突然发病死亡,为鉴定导致其发病死亡的致病菌,试验无菌采取病死猪病变肺组织,通过细菌分离培养、染色镜检、生化试验、16S rRNA PCR扩增等方法进行鉴定,并对分离菌进行药敏试验、血清型鉴定、毒素基因鉴定、耐药基因检测及毒力试验。结果显示,从病变肺中分离到1株病原菌,分离菌的菌落形态、菌体形态、生化特征和16S rRNA基因序列均与胸膜肺炎放线杆菌相符,将其命名为APP GZBJ2020;药敏试验显示,APP GZBJ2020株对头孢曲松、头孢哌酮、青霉素等9种药物敏感,对头孢氨苄和米诺环素中度敏感,对庆大霉素、红霉素、新霉素等9种药物耐药;血清型鉴定结果显示其为血清5型胸膜肺炎放线杆菌;毒素鉴定结果显示APP GZBJ2020株含有ApxⅠ、ApxⅡ及ApxⅣ三种毒素基因;耐药基因检测结果显示,APP GZBJ2020株携带有tem、gyrB、parC和parE四种耐药基因;动物毒力试验结果显示,APP GZBJ2020株对小鼠有致病性。试验成功从病死经产母猪病变肺中分离到1株血清5型胸膜肺炎放线杆菌,试验结果可为该猪场治疗发病猪提供一定帮助,并为了解胸膜肺炎放线杆菌在贵州省的流行血清型提供数据支持。
In November 2020,a sudden death of a sow occurred in a swine farm in Bijie,Guizhou,China.In order to confirm the cause of the death,the lung tissues of the dead sow was aseptically collected for identification through bacterial isolation and culture,staining and microscopy observation,biochemical tests,16S rRNA PCR amplification and other methods.Drug susceptibility test,serotype identification,toxin gene identification,drug resistance gene detection and virulence test were performed on the isolate.The results showed that one strain of pathogenic bacteria was isolated from the lung,and the colony morphology,cell morphology,biochemical characteristics and 16S rRNA gene sequence of the isolate were consistent with the infectious Actinobacillus pleuropneumoniae,which was named APP GZBJ2020.The drug sensitivity test showed that APP GZBJ2020 strain were sensitive to 9 drugsincluding ceftriaxone,cefoperazone and penicillin,moderately sensitive to cephalexin and minocyclinee,resistance to 9 drugs including gentamicin,erythromycin and neomycin.The serotype identification result showed that APP GZBJ2020 strain was serotype 5 Actinobacillus pleuropneumoniae.The toxin identification results showed that APP GZBJ2020 strain contains three virulence genes:ApxⅠ,ApxⅡand ApxⅣ.The test results of drug resistance genes showed that APP GZBJ2020 strain contains four drug resistance genes:tem,gyrB,parC and parE.Animal virulence test results show that APP GZBJ2020 strain was pathogenic to mice.This study successfully isolated a serotype 5 Actinobacillus pleuropneumoniae from the lung of a dead sow,and the results provided some help for the treatment of diseased pigs in the swine farm,and provided data for understanding the prevalent serotypes of Actinobacillus pleuropneumoniae in Guizhou Province.
