摘要
目的:通过计算机网络药理学技术分析柴胡治疗癫痫的活性化合物及其作用靶点,并预测其作用机制。方法:通过中药系统药理学分析平台筛选出柴胡的化学成分和靶点,利用GeneCards和OMIM数据库筛选出癫痫相关的靶点,基于在线软件venny 2.1.0分析获得108个柴胡与癫痫的共同靶点,并使用Cytoscape软件构建“柴胡活性化合物-靶点”网络图,STRING数据库构建蛋白互作网络(PPI网络),并进行PPI网络拓扑分析筛选出核心靶点,利用clusterProfiler对108个共同靶点进行GO分析和KEGG分析,再通过Cytoscape软件构建KEGG关系网络。结果:从数据库中筛选出17个柴胡的活性成分,对应靶点166个,包括与癫痫相关的108个靶点。对共同靶点进行PPI网络拓扑分析,筛选获得12个核心靶点。通过GO分析获得富集前20位的GO条目,KEGG分析除了获得富集前20位的通路外,还获得了与癫痫密切相关的MAPK信号通路和细胞凋亡通路等。结论:本研究初步揭示了柴胡治疗癫痫的可能作用机制,推测柴胡可能通过多成分、多靶点、多信号通路共同起到治疗癫痫的作用,为进一步深入研究提供了理论基础。
Objective:To analyze the active compounds of Chaihu(Bupleurum)for treating epilepsy and their targets by computer network pharmacology technology,and predict its mechanism of action.Methods:The chemical components and targets of Chaihu(Bupleurum)were screened out through the Traditional Chinese Medicine System Pharmacology Analysis Platform(TCMSP),and the epilepsy-related targets were screened out using GeneCards and OMIM database.Based on the online software venny 2.1.0 analysis,108 common targets of Chaihu(Bupleurum)and epilepsy were obtained,Cytoscape software was used to construct the"Chaihu(Bupleurum)active compound-target"network diagram,the STRING database was used to construct a protein interaction network(PPI network),and perform PPI network topology analysis was used to screen out core targets.ClusterProfiler was used to perform GO analysis and KEGG analysis on 108 common targets,and then built a KEGG relationship network through Cytoscape software.Results:17 active ingredients of Chaihu(Bupleurum)were screened from the database,corresponding to 166 targets,including 108 targets related to epilepsy.PPI network topology analysis was performed on common targets,and 12 core targets were screened.Through GO analysis,the top 20 enriched GO entries were obtained.In addition to the top 20 enriched pathways,KEGG analysis also obtained the MAPK signaling pathway and apoptosis pathway closely related to epilepsy.Conclusion:This study initially reveals the possible mechanism of Chaihu(Bupleurum)for treating epilepsy.It is speculated that Chaihu(Bupleurum)may play a role in treating epilepsy through multi-component,multi-target,and multi-signal pathways,which provides a theoretical basis for further in-depth research and experimental verification.
