人参皂苷RG3对自身免疫性甲状腺炎小鼠甲状腺功能和辅助型T细胞亚群的影响及相关机制 被引量:6

摘要 目的研究人参皂苷RG3治疗对自身免疫性甲状腺炎(AT)小鼠甲状腺功能以及外周血、脾脏辅助T细胞亚群的影响。方法选取30只健康无特定病原体(SPF)级BALB/C雌性小鼠作为研究对象,6月龄,体重16~18 g。随机数表法将小鼠均分为3组,即对照组(健康小鼠,n=10)、模型组(AT小鼠,n=10)和治疗组(人参皂苷Rg3治疗的AT小鼠,n=10)。通过过量碘联合猪甲状腺球蛋白诱导建立AT小鼠模型。治疗组小鼠通过灌胃给予人参皂苷RG3治疗,模型组和对照组给予等量的0.9%氯化钠溶液,连续治疗3周。酶联免疫吸附试验测定3组小鼠血清促甲状腺素(TSH)、游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)、甲状腺球蛋白抗体(TgAb)、白细胞介素-4(IL-4)和γ干扰素(IFN-γ)含量,流式细胞术检测3组小鼠CD3^(+)CD8^(+)IL-4^(+)和CD3^(+)CD8^(+)IFN-γ^(+)辅助T淋巴细胞频率。结果治疗3周后,与对照组相比,模型组小鼠血清TSH、TgAb、IL-4、IFN-γ以及外周血、脾脏CD3^(+)CD8^(+)IL-4^(+)和CD3^(+)CD8^(+)IFN-γ^(+)辅助T淋巴细胞频率均升高,而血清FT3和FT4含量降低,差异均有统计学意义(P<0.05);与模型组相比,治疗组小鼠血清TSH、TgAb、IL-4、IFN-γ以及外周血、脾脏CD3^(+)CD8^(+)IL-4^(+)和CD3^(+)CD8^(+)IFN-γ^(+)辅助T淋巴细胞频率均降低,而血清FT3和FT4含量升高,差异均有统计学意义(P<0.05)。结论人参皂苷RG3可以有效改善AT小鼠甲状腺功能,并且可以通过调控CD3^(+)CD8^(+)IL-4^(+)和CD3^(+)CD8^(+)IFN-γ^(+)辅助T淋巴细胞频率而降低IL-4和IFN-γ的分泌,降低AT小鼠自身免疫反应。 Objective To study the effect of ginsenoside RG3 on thyroid function and helper T cell subsets in autoimmune thyroiditis(AT)mice.Methods Thirty healthy SPF-free BALB/C female mice,6 months old,weighing 16-18 g were selected as the research objects.According to the random number table method,the mice were divided into 3 groups,namely the control group(healthy mice,n=10),the model group(AT mice,n=10)and the treatment group(AT mice treated with ginsenoside Rg3,n=10).AT mouse models were established by excess iodine combined with porcine thyroglobulin.Mice in the treatment group were given ginsenoside RG3 by gavage,and the model group and the control group(n=10)were given the same amount of 0.9% sodium chloride solution for 3 consecutive weeks.Enzyme-linked immunosorbent assay was used to determine serum thyroid stimulating hormone(TSH),free triiodothyronine(FT3),free thyroxine(FT4),anti-mouse thyroglobulin antibody(TgAb),interleukin-4(IL-4),and γ-interferon(IFN-γ)levels in each group of mice,and flow cytometry to detect the frequency of peripheral blood,spleen CD3^(+)CD8^(+)IL-4^(+) and CD3^(+)CD8^(+)IFN-γ^(+)helper T lymphocytes increased,in each group of mice.Results After 3 weeks treatment,compared with the control group,the model group mice serum TSH,TgAb,IL-4,IFN-γ and the frequency of peripheral blood,spleen CD3^(+)CD8^(+)IL-4^(+) and CD3^(+)CD8^(+)IFN-γ^(+) helper T lymphocytes increased,while the levels of serum FT3 and FT4 decreased,and the differences were statistically significant(P<0.05);Compared with the model group,the levels of serum TSH,TgAb,IL-4,IFN-γ,the frequency of peripheral blood,spleen CD3^(+)CD8^(+)IL-4^(+) and CD3^(+)CD8^(+)IFN-γ^(+) helper T lymphocytes in the treatment group decreased,while the levels of serum FT3 and FT4 increased,and the differences were statistically significant(P<0.05).Conclusion Ginsenoside RG3 can effectively improve the thyroid function of AT mice,and can reduce the secretion of IL-4 and IFN-γ by regulating the frequency of CD3^(+)CD8^(+)IL-4^(+) and CD3^(+)CD8^(+)IFN-γ^(+) helper T lymphocytes.Autoimmune response in AT mice.
