摘要
目的通过体外模型研究不可逆电穿孔(IRE)对不同分化程度结肠癌细胞株增殖的抑制效果及作用机理,考察IRE对抑制消化道肿瘤生长的有效性。方法脉冲发生装置设置不同电击参数,对不同分化程度的结肠癌细胞HCT116(低分化)、SW480和SW620(中分化)、LoVo(高分化)进行电击,24 h后通过MTT细胞增殖实验检测其存活率。结果MTT细胞增殖实验表明,IRE电场强度达到1500 V/cm,脉冲个数高于40时,可明显抑制中低分化结肠癌细胞株的增殖,对于高分化结肠癌细胞株LoVo细胞增殖的影响明显减弱。结论IRE对不同分化程度的结肠肿瘤细胞株具有不同程度的不可逆性杀伤作用,对中低分化肠癌效果更佳。
Objective To investigate the effect and mechanism of irreversible electroporation(IRE)on the proliferation of colon cancer cell lines with different degrees of differentiation in vitro.Methods The pulse generator was set with different shock parameters to shock HCT116(poorly differentiated),SW480 and SW620(moderately differentiated)and LoVo(well differentiated)colon cancer cells.The survival rate was detected by MTT cell proliferation assay 24 hours later.Results MTT assay showed that when IRE electric field intensity reached 1500 V/cm and the number of pulses was higher than 40,the proliferation abilities of poorly differentiated colon cancer cell line was significantly inhibited,and the effect on the proliferation of highly differentiated colon cancer cell line LoVo was significantly weakened.Conclusion IRE could irreversibly kill colon cancer cell lines with different degrees of differentiation,and has better effect on moderately and poorly differentiated colon cancer cells.
