PGRN、TLR4、TLR2在衣原体生殖道感染不孕患者中的表达及意义 被引量:2

Expression and significance of PGRN, TLR4 and TLR2 in infertile patients with chlamydia genital tract infection

摘要 目的分析颗粒蛋白前体(PGRN)、Toll样受体4(TLR4)、Toll样受体2(TLR2)在衣原体生殖道感染不孕患者中的表达及意义。方法选取2019年1月至2020年1月鄂东医疗集团黄石市中心医院收治的84例衣原体生殖道感染不孕患者作为感染组,同期选取行健康体检的50例健康志愿者作为对照组。采集阴道分泌物、血清样本,采用实时荧光定量PCR法检测阴道分泌物中PGRN、TLR4、TLR2表达水平,采用酶联免疫吸附试验法(ELISA法)检测血清中PGRN、TLR4、TLR2表达水平。对比两组PGRN、TLR4、TLR2表达变化,并分析三指标之间的相关性。结果与对照组、慢性感染、流产次数≤2次比较,感染组、急性感染、流产次数>2次患者阴道分泌物和血清中的PGRN、TLR4、TLR2表达水平均升高,差异具有统计学意义(P<0.05)。PGRN、TLR4、TLR2之间Pearson相关性分析结果显示,PGRN与TLR4之间为正相关(r=0.298,P=0.006);PGRN与TLR2之间为正相关(r=0.274,P=0.012);TLR4与TLR2之间为正相关(r=0.257,P=0.019)。结论PGRN、TLR4、TLR2在衣原体生殖道感染不孕患者中均高表达,与患者感染类型、流产次数相关,且三者之间均为正相关,可能共同参与此病的发展。 Objective To analyze the expression and significance of Progranulin(PGRN),Toll-like receptor 4(TLR4)and Toll-like receptor 2(TLR2)in infertility patients with chlamydia genital tract infection.Methods 84 infertile patients with chlamydia genital tract infection admitted to Huangshi Central Hospital of Eastern Hubei Medical Group from January 2019 to January 2020 were selected as the infection group,and 50 healthy volunteers who underwent physical examination during the same period were selected as the control group.Vaginal secretions and serum samples were collected,and the expression levels of PGRN,TLR4 and TLR2 in vaginal secretions were detected by real-time fluorescence quantitative PCR,and the expression levels of PGRN,TLR4 and TLR2 in serum were detected by enzyme linked immunosorbent assay(ELISA).The expression changes of PGRN,TLR4 and TLR2 were compared between the two groups,and the correlation between the three indexes was analyzed.Results Compared with control group,chronic infection and abortion≤2 times,the expression levels of PGRN,TLR4 and TLR2 in vaginal secretions and serum of infection group,acute infection and abortion>2 times were increased,with statistical significance(P<0.05).Pearson correlation analysis among PGRN,TLR4 and TLR2 showed that there was a positive correlation between PGRN and TLR4(r=0.298,P=0.006).PGRN was positively correlated with TLR2(r=0.274,P=0.012).There was a positive correlation between TLR4 and TLR2(r=0.257,P=0.019).Conclusions PGRN,TLR4 and TLR2 are all highly expressed in infertile patients with chlamydial genital tract infection,and are related to the type of infection and the number of miscarriages,and the three are positively correlated,and may jointly participate in the development of this disease.
