芪药消渴胶囊对胰岛素抵抗大鼠糖脂代谢和肝脏组织IRS-2蛋白表达的影响 被引量:1

Effect of Qiyao Xiaoke Capsule on IRS-2Protein Expression in Liver Tissue of Rats with Insulin Resistance

摘要 目的:研究了芪药消渴胶囊对胰岛素抵抗(IR)大鼠肝脏组织胰岛素受体底物-2(IRS-2)蛋白表达的影响。方法:选取清洁级SD健康大鼠50只,选取10只作为对照组饲喂常规饲料,其余40只饲喂高脂饲料建立IR大鼠模型,并随机分为IR模型组、低剂量组、高剂量组和二甲双胍组。低剂量组和高剂量组分别使用低剂量和高剂量芪药消渴胶囊混悬液灌胃,二甲双胍组使用二甲双胍灌胃。比较不同组别大鼠干预后的一般情况、血清学指标、肝脏和腹腔脂肪参数以及IRS-2蛋白表达水平。结果:IR模型组的体质量、INS、TG、TC、FFA和HOMA-IR水平均明显高于对照组、低剂量组、高剂量组和二甲双胍组(P<0.05)。IR模型组的肝脏重量、腹腔脂肪重量、肝脏指数和腹脂指数均明显高于对照组、低剂量组、高剂量组和二甲双胍组(P<0.05)。IR模型组IRS-2蛋白表达水平明显低于对照组、低剂量组、高剂量组和二甲双胍组(P<0.05)。结论:芪药消渴胶囊可有效改善IR大鼠糖脂代谢紊乱,减少腹腔和肝脏脂肪堆积,这可能与其能够上调肝脏组织中IRS-2蛋白表达相关。 Objective: To study the effect of Qiyao Xiaoke Capsule on IRS-2 protein expression in liver tissue of rats with insulin resistance were studied. Methods: 50 healthy SD rats of clean grade were selected, 10 were selected as a controlled group and fed with regular feed, and the other 40 were fed with high-fat feed to establish an IR rat model, and were randomly divided into IR a model group, a low-dose group, and a high-dose group and a metformin group. The low-dose group and the high-dose group were given low-dose and high-dose Qiyao Xiaoke Capsule Suspension respectively by gavage, and the Metformin group was gavage with Metformin. The general conditions, serum indexes, liver and abdominal fat parameters, and IRS-2 protein expression levels of the different groups of rats after intervention were compared. Results: The weight, INS,TG,TC,FFA and HOMA-IR levels of the IR model group were significantly higher than those of the controlled group, the low-dose group, the high-dose group and the Metformin group(P<0.05). The liver weight, abdominal fat weight, liver index and abdominal fat index of the IR model group were significantly higher than those of the controlled group, the low-dose group, the high-dose group and the Metformin group(P<0.05). The expression level of IRS-2 protein in the IR model group was significantly lower than that of the controlled group, the low-dose group, the high-dose group and the Metformin group(P<0.05). Conclusion: Qiyao Xiaoke Capsule can effectively improve the glucose and lipid metabolism disorder of IR rats and reduce the accumulation of abdominal cavity and liver fat, which may be related to its ability to up-regulate the expression of IRS-2 protein in liver tissue.
