摘要
目的:构建甘草酸水凝胶载药体系并研究其对黄芩素体外属性的影响。方法:通过甘草酸单铵盐自组装行为构建甘草酸水凝胶载药体系,并以流变学特征和黏度为指标考察其影响因素;分析甘草酸水凝胶的稳定性、流动性和触变性等性质并研究其对黄芩素载药和释放行为的影响。结果:制备甘草酸水凝胶的最佳浓度为10mg/mL,水浴温度为80℃,分散介质为水,冷却方式为低温冷却;甘草酸水凝胶室温下稳定性较好,且有触变恢复性和剪切变稀的特点;甘草酸水凝胶能作为黄芩素的载体并对黄芩素有缓释作用。结论:甘草酸单铵盐能通过自组装形成甘草酸水凝胶,该凝胶能增加黄芩素溶解度并对黄芩素有缓释的作用,因此甘草酸在一定程度上可以改变黄芩素的生物利用度。
Objective: Construction of glycyrrhizic acid hydrogel drug loading system and its effect on properties of baicalein in vitro. Methods: The glycyrrhizic acid hydrogel drug loading system was constructed by self-assembly of monoammonium glycyrrhizinate, and its influence factors were investigated by rheological characteristics and viscosity. The stability, liquidity and thixotropy of glycyrrhizic acid hydrogel were analyzed, and the effects of glycyrrhizic acid on baicalein loading and release behavior were studied. Results: The optimum concentration of glycyrrhizic acid hydrogel was 10 mg/mL, the water bath temperature was 80℃, the dispersing medium was water, and the cooling method was low temperature cooling;the glycyrrhizic acid hydrogel had good stability at room temperature, and had the characteristics of thixotropism and shear thincning;glycyrrhizic acid hydrogel could be used as the carrier of baicalein, and had a sustained-release effect on baicalein. Conclusion: Monoammonium glycyrrhizinate can form glycyrrhizic acid hydrogel through self-assembly. The gel can increase the solubility of baicalein and has a sustained-release effect on baicalein, therefore, glycyrrhizic acid hydrogel can change the bioavailability of baicalein to a certain extent.
