摘要
为提高甘草育种效率,促进优良品种的保存和开发利用,以甘草(Glycurrihiza uralensis Fisch.)无菌苗的愈伤组织为材料,通过正交试验比较不同激素组合对甘草愈伤组织再分化的影响,探究愈伤组织再分化的最适条件。结果表明:生芽最佳培养基为MS+IBA 0.4 mg·L-1+IAA 0.4 mg·L-1+KT 0.3 mg·L-1,芽数4.87个·块-1,芽长2.32 cm,诱导率达64.44%;诱导甘草不定根最佳培养基配方为MS+IBA 0.4 mg·L-1+IAA 0.4 mg·L-1+KT 0.1 mg·L-1,根数5.67条·块-1,根长3.56 cm,诱导率达55.56%。
In order to improve the breeding efficiency of Glycyrrhiza uralensis Fisch.and promote the preservation and development of fine varieties,the callus of Glycyrrhiza uralensis Fisch.was used as material,and the effects of different hormone combinations on callus redifferentiation of Glycyrrhiza uralensis Fisch were compared by orthogonal test,and the optimum conditions for callus redifferentiation were explored.The results showed that the best medium for bud growth was MS+IBA 0.4 mg·L-1+IAA 0.4 mg·L-1+KT 0.3 mg·L-1,with 4.87 buds and 2.32 cm length,with an induction rate of 64.44%.The optimal medium formula for inducing adventitious roots of Glycyrrhiza uralensis Fisch was MS+IBA 0.4 mg·L-1+IAA 0.4 mg·L-1+KT 0.1 mg·L-1,the number of roots is 5.67,the length of roots was 3.56 cm,and the induction rate was 55.56%.
