摘要
目的:探讨肿瘤抑癌基因7L(ST7L)在非小细胞肺癌(NSCLC)中的表达水平及具体分子机制。方法:运用临床数据库(TCGA和GEO)分析NSCLC临床样本中ST7L的表达水平;实时荧光定量PCR(RT-q PCR)检测25对NSCLC患者及肺癌细胞系中ST7L的表达水平;CCK8实验、集落形成实验、细胞周期实验、Hoechst试剂盒染色检测ST7L对肺癌A549和H1299细胞生长能力、细胞周期及细胞凋亡的影响;Western blot、荧光报告载体实验和免疫荧光检测ST7L对WNT/β-catenin通路的调控作用。结果:ST7L在NSCLC组织及细胞系中低表达,差异有统计学意义(P<0.05)。NSCLC患者组织中ST7L的表达水平与肿瘤大小、TNM分期及远端转移呈负相关(P<0.05),与年龄、性别及组织学分类无相关性(P>0.05)。ST7L通过抑制细胞周期进程和促进细胞凋亡抑制A549和H1299细胞的增殖能力(P<0.05)。ST7L在A549和H1299细胞中抑制WNT信号通路活化(P<0.05)。结论:ST7L通过失活WNT/β-catenin通路在NSCLC细胞中抑制细胞的恶性行为。
Objective:To investigate the expression level and specific molecular role of tumor suppressor gene 7 L(ST7L)in non-small cell lung cancer(NSCLC).Methods:Clinical databases(TCGA and GEO)were used to analyze expression level of ST7L in clinical samples of NSCLC. Real-time fluorescence quantitative PCR(RT-qPCR)was used to detect ST7L in 25 pairs of NSCLC patients and lung cancer cell lines. CCK8 test,colony formation test,cell cycle test,and Hoechst kit staining were used to detect effects of ST7L on growth ability,cell cycle,and apoptosis of lung cancer A549 and H1299 cells;Western blot,fluorescent reporter,and immunofluorescence were used to detect the regulatory effect of ST7L on WNT/β-catenin pathway.Results:ST7L was low-expressed in NSCLC tissues and its cell lines,with a statistically significant(P<0.05). Expression level of ST7L in tissues of patients with NSCLC was negatively correlated with tumor size,TNM stage,and distant metastasis(P<0.05). There was no correlation with age,gender,and histological classification(P>0.05). ST7L inhibited the proliferation of A549 and H1299 cells by inhibiting cell cycle progression and promoting apoptosis(P<0.05). ST7L inhibited activation of WNT signaling pathway in A549 and H1299 cells(P<0.05).Conclusion:ST7L can inhibit the malignant phenotype of NSCLC cells by inactivating the WNT/β-catenin signaling pathway.
