摘要 免疫已经广泛应用于骨代谢性疾病。适应性免疫的生理学功能有利于调节骨骼系统,然而在病理状态下,如类风湿性关节炎、骨关节炎、骨质疏松症等,骨骼系统会随着免疫应答做出相应调整,从而导致骨丢失或骨折风险增加等后果。T淋巴细胞和其他各种免疫细胞参与骨调节及骨稳态。本综述将讨论T细胞及其亚群等淋巴细胞在骨代谢性疾病中的作用。活化T细胞可通过分泌不同的细胞因子直接或间接调节骨健康,从而调节骨代谢。雌激素的缺乏可通过IL-7、IFN-γ、TNF等诱发T细胞增殖,引起骨丢失,导致骨质疏松。CD4+T细胞是类风湿性关节炎炎性滑膜炎形成、软骨及骨破坏的关键因素,且衰老CD4+T细胞作用更甚。CTLA4-Ig对于缓解骨质破坏具有积极作用。在骨关节炎的发病过程中,活化T细胞或异常T细胞的免疫应答可诱发关节炎中骨丢失和骨破坏。CD4+T、Th1、Th1/Th2、Th17等增多会增强破骨细胞活性,而CD8+T细胞、Treg、CTLA-4等增多则有抑制破骨细胞的作用。因此,随着免疫细胞与骨代谢之间的不断交叉,骨免疫学将越来越受到人们的重视,了解并熟悉免疫细胞与骨代谢之间的关系非常有必要。 Immune system is widely used in bone metabolic diseases. Physiology function of adaptive immune was the benefit of regulating bone system while rheumatoid arthritis,osteoarthritis and osteoporosis diseases occurred at immune disfunction that bone system will take relevant adjustment with immune response and induced bone loss or increased risk of fracture. T lymphocyte and other immune cells participated in bone regulation and remodeling. This review will discuss the role of T cells and their subpopulation in bone metabolic diseases. Activated T lymphocyte cells can modulate bone health and bone metabolism directly and indirectly through secreting several cytokines. The estrogen deficiency will initiate bone loss of osteoporosis owning to T lymphocyte cells proliferation by IL-7,IFN-γ,TNF and so on. CD4+T cells are the key factors for formation of inflammatory synovitis,destruction of cartilage and bone in rheumatoid arthritis,so did as to senescent CD4+T cells. CTLA-4 Ig has a positive function for release destruction of bone. Immune response of activated T lymphocyte cells and abnormal T cells can induce bone loss and destruction at osteoarthritis. Increased of CD4+T,Th1,Th1/Th2,Th17 etc. may promote activity of osteoclasts,while increased of CD8+T,Treg,CTLA-4 etc. can inhibit activity of osteoclasts. Thereby,the more constantly cross between immune cells and bone metabolism,the more highlight of osteoimmunology for people,and it is necessary to realize and master the relationship between immune cells and bone metabolism.
