益气化痰祛瘀法调控TLR4/MyD88通路改善慢性间歇性缺氧大鼠心肌炎症损伤 被引量:3

摘要 目的:探讨益气化痰祛瘀法对慢性间歇性缺氧(CIH)大鼠心肌损伤的作用及分子机制。方法:30只雄性SD大鼠分为Control组、Model组、TCM组,每组10只。Model组和TCM组大鼠每天9:00-17:00置于低氧舱中,制备CIH模型,Control组同一时间予间歇常氧处理。造模第1天起TCM组给予益气化痰祛瘀方38.4 ml/(kg·d)灌胃,余两组予2 ml生理盐水灌胃,6周后检测CK、LDH,Western Blot(WB)、RT-PCR检测TLR4、MyD88表达水平,ELISA检测MDA含量。结果:与Model组比,TCM组的CK、LDH、MDA含量均较低(P<0.05),且TLR4、MyD88表达明显下调(P<0.05)。结论:益气化痰祛瘀法减少CIH大鼠心肌炎症因子表达,改善心功能,可能与抑制TLR4/MyD88信号通路诱导的炎症反应有关。 Objective:To investigate the effect and molecular mechanism of Yiqi Huatan Quyu method on myocardial injury in chronic intermittent hypoxia (CIH) rats.Method:Thirty male SD rats were divided into Control group,Model group and TCM group with 10 rats in each group.Rats in Model group and TCM group were placed in hypoxic chamber to prepare CIH model from 9:00 to 17:00 every day.Rats in Control group were given intermittent normoxia treatment at the same time.TCM group was given Yiqi Huatan Quyu recipe 38.4 ml/(kg·d) by gavage,and the other groups were given 2 ml normal saline by gavage from the first day.After 6 weeks,the levels of CK,LDH were detected.TLR4 and MyD88 were detected by Western Blot (WB) and RT-PCR.The content of MDA was detected by ELISA.Result:Compared with Model group,the contents of CK,LDH and MDA were decreased in TCM group (P<0.05),and the expressions of TLR4 and MyD88 were significantly down regulated (P<0.05).Conclusion:Yiqi Huatan Quyu method can reduce the expression of myocarditis factors and improve cardiac function in CIH rats.It may be related to the inhibition of inflammatory response induced by TLR4/MyD88 signaling pathway.
