摘要
目的建立银菊汤中三种成分的含量同时测定方法,评价该汤剂的质量。方法使用高效液相色谱法,Agilent Eclipse C 18色谱柱(250×4.6 mm,粒径5μm),乙腈(A)-0.2%磷酸水溶液(B)为流动相,梯度程序为:0~15 min,10%~13%A;15~25 min,13%~25%A;25-35 min,25%~75%A;35~45 mi n,75%~10%A;流速1.0 mL·min^(-1),检测波长300 nm,柱温25℃,进样量10μL,检测指标为绿原酸、黄芩苷、连翘苷,平行制备10份样品进行含量测定。结果根据10批供试品溶液测定结果计算,绿原酸含量为0.5904~0.7780 mg·mL^(-1),黄芩苷含量为0.3654~0.5836 mg·mL^(-1),连翘苷含量为0.1573~0.2957 mg·mL^(-1)。结论本研究所用方法快速、准确,能够较为科学、全面地评价银菊汤的质量。
OBJECTIVE To evaluate the quality of YinJu Decoction,by establishing a simultaneous determination method for three compounds in it.METHODS Determination used high performance liquid chromatography(HPLC)with Agilent Eclipse C18 column(250×4.6 mm,particle size 5μm),and a mobile phase containing acetonitrile(A)-0.2%formic acid(B),the gradient program was:0~15 min,10%~13%A;15~25 min,13%~25%A;25~35 min,25%~75%A;35~45 min,75%~10%A;the flow rate was 1.0 mL·min-1,detection wavelength was 300 nm,column temperature was 25℃,injection volume was 10μL,indexes for determination was chlorogenic acid,baicalin,forsythin,and 10 batches of samples were prepared.RESULTS Calculated from the analytical results of the 10 batches of samples,chlorogenic acid contents were 0.5904~0.7780 mg·mL^(-1),baicalin contents were 0.3654-0.5836 mg·mL^(-1),forsythin contents were 0.1573~0.2957 mg·mL^(-1),respectively.CONCLUSION The method used in this study was fast and accurate,which was capable of scientific and comprehensive evaluation of YinJu Decoction′s quality.
