温针灸对膝骨关节炎大鼠软骨组织ADAMTS-4及MMPs-3的影响 被引量:7

Eff ect of warm acupuncture and moxibustion on the ADAMTS-4 and MMPS-3 in the cartilage tissue of the rats with knee osteoarthritis

摘要 目的观察温针灸对膝骨关节炎大鼠软骨组织ADAMTS-4、MMPs-3的影响。方法将40只大鼠采用数字随机分组法,分为正常组(10只),造模组(30只)。采用膝关节腔内注射木瓜蛋白酶造模,连续注射2周木瓜蛋白酶。对模型进行鉴定,将膝骨关节炎(KOA)造模成功的大鼠,分为造模组、温针灸组、针刺组。温针灸组给予针刺“后三里”和“膝前穴”,并在针柄处放置艾柱,治疗10 d;针刺组仅给予针刺处理,方法同温针灸组,其他组不做处理。用ELISA法检测关节液中IL-6、TNF-α含量;Western blot检测滑膜组织中ADAMTS-4、MMPs-3蛋白表达;RT-PCR检测滑膜组织中ADAMTS-4,MMPs-3的mRNA。结果造模组IL-6、TNF-α、ADAMTS-4、MMPs-3蛋白及mRNA相对含量均高于正常组(P<0.05);温针灸组各项指标均低于造模组(P<0.05)。结论温针灸能改善膝骨关节炎动物模型的病变程度,可能与抑制ADAMTS-4、MMPs-3表达有关。 Objective To observe the eff ect of warm acupuncture and moxibustion on the ADAMTS-4 and MMPs-3 in the cartilage tissue of the rats with knee osteoarthritis(KOA).Methods A total of forty rats were divided into a normal group(10 rats)and a model group(30 rats)by the digital random grouping method.Papain was continuously injected into the knee joint cavity for 2 weeks to create the model.The model was identifi ed,and the rats successfully modeled for knee osteoarthritis were randomly divided into the model model,warm acupuncture and moxibustion group,and acupuncture group.The warm acupuncture and moxibustion group was given acupuncture on Housanli and Xiqian points,with moxa retained at the needle handles,and the treatment lasted for 10 days.The acupuncture group was only given the acupuncture treatment,with the same method,and the other groups were not treated.The content of IL-6 and TNF-αin the synovial fl uid were detected by the ELISA method.The protein expression of ADAMTS-4 and MMPs-3 in the synovial tissue was detected by Western blot.The mRNA level of ADAMTS-4 and MMPs-3 in the synovial tissue was detected by RT-PCR.Results IL-6 and TNF-α,and the relative contents of ADAMTS-4,MMPs-3 protein and mRNA in the model group were all significantly higher than those in the normal group(P<0.05).The indexes in the warm acupuncture and moxibustion group were lower than those in the model group(P<0.05).Conclusion Warm acupuncture and moxibustion can improve the degree of lesions in the animal model of knee osteoarthritis,which may be related to the inhibition of the ADAMTS-4 and MMPs-3 expression.
