摘要
质子泵抑制剂(PPIs)通过作用于胃H^(+),K^(+)-ATP酶,抑制胃酸分泌,其因具有强大的抑酸作用而成为消化性酸相关疾病治疗和预防的首选药物。PPIs在抗嗜酸细胞性食管炎、抗胰腺炎、抑制口腔细菌、抗结核分枝杆菌、抗真菌等方面具有药理活性。其能通过调节特定信号通路,作用于酸相关或无关的蛋白靶点,控制肿瘤pH酸性,抑制肿瘤的发生、侵袭、转移。然而,PPIs并非理想药物。其长期使用可能引起骨质疏松、肠道菌群改变、心血管风险等不良反应。因此,开展这类药物的药理毒理评价对于规范临床用药,减少药源性不良反应,拓宽新药开发路径具有重要意义。
Proton pump inhibitors(PPIs)inhibit gastric acid secretion by acting on gastric H^(+),K^(+)-ATP enzymes,which are used as the first choice for the treatment and prevention of digestive acid related diseases.PPIs have pharmacological activities in anti-eosinophilic esophagitis,anti-pancreatitis,inhibition of oral bacteria,anti-tuberculosis and anti-fungi and so on.By regulating specific signal pathways and acting on acid-related or irrelevant protein targets,PPIs can control tumor pH acidity,and inhibit tumor occurrence,invasion and metastasis.However,PPIs are not ideal drugs,because the long term use may cause osteoporosis,intestinal flora changes,cardiovascular risk and other adverse reactions.Therefore,it is of great significance to carry out pharmacological and toxicological evaluation of these drugs for standardizing clinical medication,reducing drug-induced adverse reactions and broadening the path of new drug development.
