硫酸新霉素废水活性污泥的微生物群落结构解析 被引量:2

摘要 对硫酸新霉素废水处理中的好氧、厌氧活性污泥进行16S V3-V4扩增区域的微生物分类测序,采用宏基因组技术解析活性污泥的微生物群落结构。结果表明,从门水平而言,好氧活性污泥中的微生物主要为相对丰度为34.3%的变形菌门和相对丰度为11.5%的拟杆菌门,厌氧活性污泥中的微生物主要为相对丰度为42.7%的厚壁菌门、相对丰度为15.5%的拟杆菌门和相对丰度为10.7%的变形菌门;从属水平而言,好氧活性污泥中高丰度菌属的排序为:Parcubacteria_genera_incertae_sedis>硝化螺菌属(Nitrospira,大致与Aquihabitans相当),厌氧活性污泥中高丰度菌属的排序为:芽孢八叠球菌属(Sporosarcina)>甲烷丝菌属(Methanothrix)>泰氏菌属(Tissierella)>嗜碱菌属(Alkaliphilus)>副球菌属(Paracoccus)>Pusillimonas(大致与Petrimonas相当)>Pelolinea;通过PICRUST功能预测分析,推测好氧活性污泥中高丰度微生物的主要功能为硝化作用,厌氧活性污泥中高丰度微生物的主要功能为产甲烷;通过比较分析2种活性污泥微生物群落的主要差异物种,发现好氧活性污泥中高丰度变形菌门可能存在3种变形菌属,分别为α-变形菌、丙型变形菌、β-变形菌,厌氧活性污泥中高丰度厚壁菌门可能存在2种厚壁菌属,分别是芽孢杆菌、梭菌。该研究为活性污泥A/O法处理硫酸新霉素废水奠定了基础。 We carried out microbial classification and sequencing in 16S V3-V4 amplification region of aerobic and anaerobic activated sludge collected in the treatment of Neomycin Sulfate wastewater,and analyzed the microbial community structure of activated sludge by metagenomic technology.The results show that,at phylum level,the main microorganisms in aerobic activated sludge are Proteobacteria and Bacteroidetes with relative abundances of 34.3%and 11.5%,respectively;the main microorganisms in anaerobic activated sludge are Firmicutes,Bacteroidetes,and Proteobacteria with relative abundances of 42.7%,15.5%,and 10.7%,respectively.At genus level,the order of high abundance bacteria in aerobic activated sludge is:Parcubacteria_genera_incertae_sedis>Nitrospira(approximately equivalent to Aquihabitans);the order of high abundance bacteria in anaerobic activated sludge is:Sporosarcina>Methanothrix>Tissierella>Alkaliphilus>Paracoccus>Pusillimonas(approximately equivalent to Petrimonas)>Pelolinea.Through PICRUST functional prediction analysis,it is inferred that the main function of the microorganisms with higher abundances in aerobic activated sludge is nitrification,and the main function of the microorganisms with higher abundances in anaerobic activated sludge is methanogenesis.According to the analysis of the main differential species between the two kinds of activated sludge,the Proteobacteria with the highest abundance in aerobic activated sludge may contain three kinds of Proteobacteria with Alphaproteobacteria,Gammaproteobacteria,and Betaproteobacteria,while the Firmicutes with the highest abundance in anaerobic activated sludge may contain two kinds of Firmicutes with Bacilli and Clostridia.This study lays a foundation for the treatment of Neomycin Sulfate wastewater by activated sludge A/O process.
