摘要
目的探究机体的代谢综合征和血清睾酮水平对SD大鼠前列腺组织增生的影响,并在大鼠前列腺增生模型中探索Ⅱ型5α还原酶(SRD5A2)的表达水平及意义。方法运用高脂高糖饮食模式,建立代谢综合征合并前列腺增生大鼠模型,建立手术去势处理,实验组予以皮下注射睾酮,获取第40天SD大鼠的体质量、前列腺组织重量、前列腺指数等参数,并采用免疫组织化学染色和实时定量PCR方法检测大鼠前列腺组织样本中SRD5A2表达情况,并分析其与前列腺指数的影响关系。结果高脂高糖饮食饲养组可表现出机体代谢综合征症状,较普通饮食饲养组更易发生前列腺组织增生。手术去势处理后皮下注射睾酮可诱发前列腺明显增生。SRD5A2在高脂高糖饲养组的表达水平显著高于普通饲养组(P<0.05)。结论代谢综合征和睾酮均是大鼠前列腺组织增生的重要影响因素,SRD5A2在前列腺组织内的表达量与前列腺增生程度呈正相关性。
Objective To explore the effects of metabolic syndrome and serum testosterone on prostatic tissue hyperplasia in SD rats,and explore the expression and significance ofⅡtype 5αreductive-SRD5A2 in the rat model of prostatic hyperplasia.Methods A rat model of metabolic syndrome complicated with prostatic hyperplasia was established by high fat and high sugar diet.The experimental group was given subcutaneous injection of testosterone.The body weight,prostate tissue weight,prostate index and other parameters of SD rats were obtained after 40 days.Immunohistochemical staining and real-time quantitative PCR were used to detect the expression of SRD5A2 in rat prostate tissue samples,and the relationship between SRD5A2 expression and prostate index was analyzed.Results The group fed with high fat and high sugar diet showed the symptoms of metabolic syndrome and was more prone to prostate tissue hyperplasia than the group fed with ordinary diet.Prostatic hyperplasia can be induced by subcutaneous injection of testosterone after surgical castration.The expression level of SRD5A2 in high fat and high sugar feeding group was significantly higher than that in ordinary feeding group(P<0.05).Conclusions The metabolic syndrome and testosterone are important influencing factors for prostate tissue hyperplasia in rats.The expression of SRD5A2 in prostate is positively correlated with the degree of prostate hyperplasia.
