摘要
目的采用超高效液相色谱-四级杆-静电轨道阱高分辨质谱(UHPLC-Q-exactive obitriap MS)对延胡索醋制前后的差异成分进行鉴别和分析。方法采用Hypersil GOLD C_(18)(2.1 mm×100 mm,1.9 μm)色谱柱,以乙腈为流动相A,0.2%氨水-5 mmol·L^(-1)甲酸铵溶液为流动相B,梯度洗脱;柱温40℃;流速0.4 mL·min^(-1);进样量2 μL;高分辨质谱采用电喷雾电离源,正、负离子模式扫描,扫描范围为50~750 Da,二级质谱碰撞能量分别为20、40和60 EV。采用Compound Discoverer 2.1软件对化学成分进行鉴别,并应用多维统计学分析延胡索醋制前后化学成分含量的变化。结果 延胡索醋制前后共有65个化学成分发生明显变化,共鉴定出其中53个化合物,除L-(+)-精氨酸、D-L-色氨酸、N-α-乙酰-L-精氨酸和sanjoinenine外,其余均为生物碱;延胡索醋制后17个化合物含量显著降低(P<0.05),48个化合物含量显著升高(P<0.05)。结论 延胡索醋制后一些化学成分的含量和比例产生明显的变化,从而增强延胡索在某些方面的临床疗效。
Objective To identify and analyze different components in Corydalis Rhizoma before and after vinegar preparation by ultra high performance liquid chromatography(UHPLC)-Q-exactive obitriap MS.Methods UHPLC was performed on Hypersil GOLD C18(2.1 mm×100 mm,1.9μm)column with acetonitrile as the mobile phase A and 0.2%ammonia water-5 mmol·L^(-1) ammonium formate solution as the mobile phase B with gradient elution.The column temperature was 40℃,the flow rate was 0.4 mL·min^(-1),and the injection volume was 2μL.The high resolution mass spectrometry adopted electrospray ionization source,with positive and negative ion mode scanning at 50~750 Da.The collision energies of the secondary mass spectra were 20,40 and 60 EV,respectively.Compound discoverer 2.1 software was used to identify the chemical components,and multi-dimensional statistical analysis was used to analyze the change in chemical components Corydalis Rhizoma before and after vinegar processing.Results Totally 65 chemical components in Corydalis Rhizoma changed significantly before and after vinegar processing and 53 compounds were identified.Except L-(+)-arginine,D-L-tryptophan,N-α-acetyl-L-arginine and sanjoinenine,the rest were alkaloids.The content of 17 compounds decreased significantly(P<0.05),while that of 48 compounds increased significantly(P<0.05).Conclusion The content and proportion of some chemical components in Corydalis Rhizoma change significantly after processing with vinegar.
