摘要
目的研究马里苷对2型糖尿病db/db小鼠心肌细胞损伤的保护作用及作用机制。方法 24只db/db小鼠随机分为模型组、阳性对照组(盐酸二甲双胍,280 mg·kg^(-1))和实验组(马里苷,50 mg·kg^(-1)),另取6只db/m小鼠作为正常对照组。每周检测小鼠体质量,检测第0、4、6、8周各组小鼠空腹血糖;药物干预8周后,检测总胆固醇、三酰甘油、空腹胰岛素,心肌组织自噬相关蛋白(LC3-Ⅱ/Ⅰ和p62)的表达水平,观察心肌组织病理变化,心肌超微结构和自噬小体。结果与正常组比较,模型组心肌纤维肥大、肿胀、排列紊乱、细胞间质增多,心肌组织p62蛋白表达水平显著升高,LC3-Ⅱ/Ⅰ显著降低,自噬小体数量减少,表明自噬被抑制,心肌组织损伤。马里苷干预后心肌纤维结构清晰、排列整齐而紧密、无明显肥大、细胞间质少,心肌组织p62蛋白表达水平显著降低,LC3-Ⅱ/Ⅰ显著升高,自噬小体数量增加,促进自噬,发挥心脏保护作用。结论马里苷通过调控自噬保护db/db小鼠的心肌损伤。
Objective To determine the protective effect and mechanism of marein on the myocardial cell injury in mice with type 2 diabetic db/db.Methods Totally 24 db/db mice were randomly divided into a model group,a metformin group(280 mg·kg^(-1))and a marein group(50 mg·kg^(-1))respectively.Six db/m mice were chosen as the normal control group.The body mass of the mice was measured weekly and the fasting blood glucose of the mice in each group was measured at 0,4,6 and 8 weeks.After 8 weeks of drug administration,all mice group were tested for serum biochemical indicators,including total cholesterol,triglyceride and fasting insulin and protein expression of autophagy related protein LC3-Ⅱ/Ⅰand p62.The pathological changes of myocardial tissue,myocardial ultrastructure and autophagomes were observed.Results Compared with the db/m normal group,the model group showed myocardial fiber hypertrophy,swelling,disordered arrangement and increased intercellular substances;the expression of p62 proteins was significantly increased,and the expression of LC3-Ⅱ/Ⅰlevel was reduced.The number of autophagosomes was decreased,indicating inhibited autophagy and damaged myocardial tissue.After the intervention of marein,the myocardial fiber structure was clear,and the arrangement was neat and tight,without obvious hypertrophy.The intercellular substance was small,the expression of p62 proteins was significantly reduced.While the LC3-Ⅱ/Ⅰand the number of autophagosomes increased,which indicated that marein promoted autophagy and played a protective role in the heart.Conclusion Marein may protect against myocardial injury in db/db mice by regulating the autophagy.
