基于网络药理学探讨补肾益气方治疗支气管哮喘的作用机制 被引量:7

Mechanism of Bushen Yiqi Formula on Bronchial Asthma Based on Network Pharmacology

摘要 目的采用网络药理学方法探讨补肾益气方治疗支气管哮喘的物质基础和作用机制。方法采用中药系统药理数据库和分析平台(TCMSP)结合文献检索获取补肾益气方的有效成分,并结合UniProt知识库得到对应的靶点信息。利用OMIM、DrugBank、TTD和Genecards数据库挖掘哮喘相关靶点。通过Cytoscape3.7.1软件构建补肾益气方有效成分-靶点及哮喘-靶点网络,绘制维恩图以获得重叠靶点。利用String平台构建重叠靶点的蛋白相互作用网络,通过Cytoscape的Cytohubba模块依据度值获得核心靶点。使用Metascape数据库,对所获靶点进行富集分析。结果筛选到黄芪17种有效成分及209个潜在靶点,淫羊藿23种有效成分及222个潜在靶点,生地黄2种有效成分及31个潜在靶点,综合分析得到237个补肾益气方有效成分的相关靶点,并同时筛选到930个哮喘相关靶点,匹配到103个重叠靶点。通过生物信息学分析,定位到20个核心靶点,进一步富集分析发现其发挥作用可能涉及白细胞介素-17(IL-17)、肿瘤坏死因子(TNF)、Toll样受体、NF-κB、NOD样受体、MAPK、PI3K-Akt等信号通路。结论补肾益气方中槲皮素、山柰酚、刺芒柄花素、淫羊藿苷、淫羊藿素、谷甾醇、豆甾醇等成分可能是治疗哮喘的物质基础,与调节炎症反应、T细胞分化、氧化应激、平滑肌细胞增殖等机制有关。 Objective To observe the material basis and mechanism of Bushen Yiqi Formula(BSYQF)in treatment of bronchial asthma by network pharmacology. Methods The effective components of BSYQF were obtained from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP) and literature retrieval,and corresponding targets were extracted from UniProt database. Asthma-associated targets were obtained from OMIM,DrugBank,TTD,and Genecards databases. The effective components of BSYQF-target networks and asthma-target networks were constructed by Cytoscape3.7.1 Software,and the Venn diagram was drawn to obtain overlapping targets. Use the String platform to construct a protein-protein interaction network of overlapping targets. Core targets were obtained via Cytohubba of Cytoscape according to the degree. Finally,the Metascape database was used to enrich Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)for the core targets. Results The results showed that 17 effective components and 209 potential targets of Radix Astragali,23 effective components and 222 potential targets of Epimedium Herb,and 2 effective components and 31 potential targets of Radix Rehmanniae. Through comprehensive analyses,237 effective components of BSYQF-associated targets were obtained. With the screening to 930 asthma-associated targets at the same time,103 overlapping targets were tracked. Using bioinformatics analyses,20 core targets were focused,and further enrichment analysis revealed that their roles might be involved in IL-17,TNF,Toll-like receptor,NF-Kappa B,NOD receptor,mitogen-activated protein kinase(MAPK),PI3 K-Akt signaling pathways. Conclusion Quercetin,kaempferol,formononetin,icariin,icaritin,sitosterol,and stigmasterol might be the material bases in BSYQF for treatment of asthma,which was related to regulating inflammatory response,T cell differentiation,oxidative stress,smooth muscle cell proliferation,and so on.
