思南黄牛的群体遗传进化分析 被引量:2

Population genetic evolution analysis of Sinan yellow cattle

摘要 为了增加对思南黄牛群体遗传多样性和种群结构的了解,试验采集12~18月龄的42头思南黄牛、5头西门塔尔牛及7头思南黄牛与西门塔尔牛杂交牛的外周血液样品,采用基因分型测序(GBS)技术对思南黄牛群体单核苷酸多态性(SNP)特征进行分析,根据SNP特征进行种群结构分析、群体进化分析、主成分分析(PCA)和群体选择分析。结果表明:获得思南黄牛群体高质量SNP位点441548个,平均SNP密度为12.338个/Mb。种群结构分析将54个样本分为A、B、C 3个集群,集群C和B亲缘关系较近;基于SNP的PCA分类结果和群体结构分析结果完全一致;群体选择分析发现3-磷酸-5-磷酸硫酸酯合成酶2(3′-phosphoadenosine 5′-phosphosulfate synthase,PAPSS2)、表皮生长因子样重复序列和盘状蛋白Ⅰ类结构域3(EGF like repeats and discoid in domains 3,EDIL3)、ADP核糖基化因子类GTPase 13B(ADP ribosylation factor like GTPase 13B,ARL13B)、突触融合蛋白19(syntaxin 19,STX19)、酪氨酸激酶2受体酪氨酸激酶4(erb-b2 receptor tyrosine kinase 4,ERBB4)、JARID2(jumonji and AT-rich interaction domain containing 2)和基质相互作用分子2(stromal interaction molecule 2,STIM2)基因受到选择,其中PAPSS2、ARL13B、STX19、EDIL3基因与牛骨骼生长有关,ERBB4、JARID2、STIM2基因与肌肉发育有关,ERBB4基因与毛色选择有关。说明不同集群牛体型相异,可能与其受选择基因偏好相关。 In order to better understand the genetic diversity and population structure of Sinan yellow cattle,peripheral blood samples were collected from forty-two 12 to 18-month old Sinan yellow cattle,five 12 to 18-month old Simmental and seven 12 to 18-month old hybrid cattle of Sinan yellow cattle and Simmental.Genotyping sequencing(GBS)was used to analyze the characteristics of single nucleotide polymorphism(SNP)in Sinan yellow cattle population.Based on SNP characteristics,population structure analysis,population evolution analysis,principal component analysis(PCA)and population selection analysis were carried out.The results showed that a total of 441548 high-quality SNP loci were obtained in Sinan yellow cattle population,the average SNP density was 12.338 SNP loci/Mb.According to the analysis result of population structure,54 samples were divided into A,B and C clusters.The cluster C and cluster B were closely related.The results of principal component analysis based on SNP were consistent with those of structure analysis.Seven genes,including 3′-phosphoadenosine 5′-phosphosulfate synthase(PAPSS2),EGF like repeats and discoid in domains 3(EDIL3),ADP ribosylation factor like GTPase 13 B(ARL13 B),syntaxin 19(STX19),erb-b2 receptor tyrosine kinase 4(ERBB4),jumonji and AT-rich interaction domain containing 2(JARID2),stromal interaction molecule 2(STIM2),were screened by population selection analysis.These genes were involved in bone development(PAPSS2,ARL13 B,STX19,EDIL3),muscle growth(ERBB4,JARID2,STIM2)and hair color selection(ERBB4).The results indicated that the body types of cattle in different groups were different,which might be related to the preference of selected genes.
