摘要
为了优化黑曲霉固态发酵喷浆玉米皮的发酵工艺,试验采用单因素试验研究底物初始含水量、发酵温度、黑曲霉接种量、发酵时间4个主要因素对黑曲霉固态发酵喷浆玉米皮效果的影响;根据响应面基本原理,利用Box-Behnken设计对发酵条件进行优化并验证;测定分析最优发酵条件发酵喷浆玉米皮前后纤维素、半纤维素、木质素和还原糖的含量变化。结果表明:理论上确定的最佳发酵工艺参数为底物初始含水量为62.5%、发酵温度为30℃、黑曲霉接种量为15%、发酵时间为72 h,此条件下得到的发酵产物的滤纸酶活性为115.389 U/g,还原糖含量为8.19%,纤维素含量由发酵前的15.75%下降到10.81%,半纤维素含量由发酵前的42.89%下降到26.05%。说明试验成功优化了黑曲霉固态发酵喷浆玉米皮的发酵工艺。
In order to optimize the fermentation process of Aspergillus niger solid-state fermentation of sprayed corn bran,a single factor experiment was conducted to study the effects of four main factors,including initial moisture content of substrate,fermentation temperature,Aspergillus niger inoculation quantity and fermentation time,on the fermentation effect of Aspergillus niger solid-state fermentation of sprayed corn bran.According to the basic principle of response surface,the fermentation conditions were optimized and verified by Box-Behnken design.The content changes of cellulose,hemicelluloses,lignin,reducing sugar of sprayed corn bran before and after fermentation under the optimal conditions were determined and analysed.The results showed that the theoretical optimum fermentation parameters were as follows:initial moisture content of the substrate was 62.5%,fermentation temperature was 30℃,Aspergillus niger inoculation quantity was 15%and fermentation time was 72 h.Under these conditions,the enzyme activity of the filter paper obtained from the fermentation product was 115.39 U/g,the mass fraction of reducing sugar was 8.19%,the content of cellulose decreased from 15.75%in before fermentation to 10.81%,and the content of hemicellulose decreased from 42.89%to 26.05%.It indicated that the fermentation process of Aspergillus niger solid-state fermentation of sprayed corn bran was successfully optimized.
