摘要
目的比较4种不同纯化方法对移植脂肪组织的影响及其体内成活率。方法将抽吸获取的脂肪组织分成4组:静置沉淀组(A组)、清洗沉淀组(B组)、离心组(C组)和棉垫吸附浓缩组(D组),并给予不同处理。体外检测不同处理组脂肪组织含水量、油脂含量、脂肪细胞完整性、基质血管成分细胞(SVF)数量及亚群、甘油-3-磷酸脱氢酶(G3PDH)活性等差异,体内评估不同处理组脂肪组织的移植效果。结果C组和D组的组织含水量小于A组和B组;D组的油脂含量小于其余各组;组织学显示4组脂肪细胞完整性无明显差别;C组与D组的SVF数量大于A组;流式细胞分析提示,D组血源性细胞较少、脂肪干细胞较多;C组和D组的G3PDH活性大于A组和B组;动物体内研究结果显示,D组脂肪重量及周脂素蛋白(PLIN)阳性细胞数明显高于其他3组。结论棉垫吸附浓缩法处理的脂肪纯度高、活性保持佳、体内存活率高。
Objective To compare the effects of four different processing methods on transplanted adipose tissue and their survival rate in vivo.Methods Adipose tissue was prepared in four groups by four different processing methods as gravity sedimentation(group A),washing and gravity sedimentation(group B),centrifugation(group C),and cotton pad concentration(group D).The water content,oil content,adipocyte integrity,number of stromal vascular component cells(SVF),ADSCs and blood-derived cells,and the activity of glycerol-3-phosphate dehydrogenase(G3PDH)were detected in vitro,and the effects of fat grafting were evaluated in vivo.Results Water content in group C and D was lower than that in group A and B;Oil content in group D was lower than that in the other groups;Histology observation showed that there was no significant difference in the adipocyte integrity among the four groups;The number of SVF in group C and group D was more than that in group A;Flow cytometry showed that there were fewer blood-derived cells and more adipose stem cells in group D;G3PDH activity in group C and D was higher than that in group A and B.Animal experiments showed that the weight and the number of Perilipin(PLIN)positive cells in group D were significantly higher than those in the other three groups.Conclusion The fat treated by cotton pad concentration has high purity,good activity and high survival rate.
