摘要
目的比较沉默调节因子1(SIRT1)在三种不同病理类型腮腺肿瘤组织中的表达水平。方法收集59例不同病理分型的口腔腮腺腺淋巴瘤、多形性腺瘤、腮腺鳞状细胞癌患者术后组织标本,分别采用RT-PCR和免疫组化技术检测SIRT1蛋白及mRNA表达水平。结果SIRT1在瘤旁组织、腮腺腺淋巴瘤(良性)、腮腺多形性腺瘤(交界性肿瘤)、腮腺鳞状细胞癌(恶性)组织中阳性率分别为16.7%、31.6%、36.3%和55.6%,差异具有统计学意义(P<0.05);SIRT1在腮腺鳞状细胞癌中高于腺淋巴瘤组织及腮腺多形性腺瘤组织中的表达水平(P<0.05),在腺淋巴瘤和腮腺多形性腺瘤之间表达差异无统计学意义(P>0.05)。结论SIRT1在唾液腺肿瘤组织中高表达,且与肿瘤恶性程度相一致,有可能作为口腔腮腺肿瘤进展的潜在标志物及治疗靶点。
Objective To investigate the expression of SIRT1 in three different pathological types of parotid gland tumors.Methods 59 cases of oral parotid adenolymphoma,pleomorphic adenoma and parotid squamous cell carcinoma were collected.The expression of SIRT1 gene in lymphoma,pleomorphic adenoma and squamous cell carcinoma of parotid gland was detected by immunohistochemistry and RT-PCR.Results The positive expression rates of SIRT1 were 16.7%,31.6%,36.3%and 55.6%in normal tissues,pleomorphic adenoma,parotid lymphoma and parotid squamous cell carcinoma(P<0.05).The expression level of SIRT1 in squamous cell carcinoma of parotid gland was higher than that in adenolymphoma and pleomorphic parotid gland and the expression of SIRT1 between in adenolymphoma and pleomorphic adenoma of parotid gland had no difference(P>0.05).Conclusion SIRT1 is highly expressed in oral tumors,which is consistent with the malignant degree of tumors.
