An age estimation method based on DNA methylation detection in the field of forensic science

摘要 目的建立一种适用于中国人群的与年龄相关的多重甲基化位点检测体系。方法采用高通量测序技术对33个文献报道的与年龄相关甲基化位点进行筛选,确定了4个在中国人群中甲基化程度与年龄高度相关的甲基化基因位点,建立了OLS多元线性回归模型数据库,可以自动计算得出甲基化程度改变与年龄相关的评估值。结果本检测体系适用于刑事案件现场常见的血斑、唾液斑等生物物证的检验鉴定,灵敏度达100ng,年龄预测准确率平均误差在3.685岁以内。结论本研究建立的多重甲基化检测体系可作为法医学应用中年龄推断的一种可靠、有效的方法,为案件侦破提供更多线索,缩小侦查范围,有助于案件的快速侦破。 Objective To establish an age-related detection system for multiple methylation sites in Chinese population. Methods We investigated methylation levels at 33 age-associated methylation sites previously reported using the next generation sequencing technology, of which 4 sites were identified. OLS multivariable linear regression model was established, and the predict age could be automatically calculated according to the degree of methylation deviation. Results The age estimation method was suitable for the identification of biological samples such as bloodstain and saliva stain, which were commonly encountered at the scene of criminal cases. The sensitivity reached 100 ng, and the mean absolute deviation(MAD) was within 3.685 years. Conclusion The multi-methylation detection system in the study could be used as a reliable and effective method for age estimation in forensic application. It could provide more clues for the criminal investigation, and help to solve cases.
