摘要
目的研究黄芩苷对心肌梗死模型大鼠心肌细胞及其免疫功能的影响。方法随机将24只SD大鼠分为假手术组、模型组与黄芩苷组,每组8只。假手术组采用丝线穿至左前降支根部,但不进行结扎;模型组和黄芩苷组采用丝线结扎左前降支。造模当天开始灌胃,1次/d,持续30 d,其中假手术组和模型组给予等量生理盐水灌胃,黄芩苷组给予100 mg/kg黄芩苷。比较三组大鼠的心功能指标心率(HR)、左心室舒张末压(LVEDP)及左心室收缩压(LVSP)、心肌组织形态学、细胞凋亡率、蛋白酶激活受体2(PAR-2)、B淋巴细胞瘤-2基因(Bcl-2)、Bcl-2相关X(Bax)蛋白的表达水平、免疫功能指标T淋巴细胞亚群(CD3^(+)、CD4^(+)、CD8^(+)、CD4^(+)/CD8^(+))变化情况。结果与假手术组比较,模型组大鼠心功能指标LVSP、Bcl-2水平降低(P<0.05),LVEDP、心肌组织蛋白Bax、PAR-2水平升高(P<0.05);与模型组比较,黄芩苷组大鼠心功能指标LVSP、Bcl-2水平升高(P<0.05),LVEDP、Bax、PAR-2水平降低(P<0.05)。与假手术组比较,模型组大鼠CD3^(+)、CD4^(+)、CD8^(+)均明显提高(P<0.05),而CD4^(+)/CD8^(+)明显降低(P<0.05);与模型组比较,黄芩苷组CD3^(+)、CD8^(+)降低(P<0.05),CD4/CD8提高(P<0.05),CD4^(+)无明显变化(P>0.05)。与假手术组比较,模型组大鼠鼠心肌细胞凋亡率升高(P<0.05);与模型组比较,黄芩苷组大鼠心肌细胞凋亡率明显降低(P<0.05)。结论黄芩苷有助于改善心肌梗死大鼠心功能,改善心肌组织的免疫功能,降低心肌细胞凋亡率,可能机制为上调Bcl-2、PAR-2蛋白水平,下调Bax蛋白表达。
Objective To study the effects of baicalin on myocardial cells and immune function in rats with myocardial infarction.Methods Twenty-four SD rats were randomly divided into sham operation group,model group and baicalin group,with 8 rats in each group.In sham operation group,silk thread was threaded to the root of the left anterior descending branch without ligation.The rats in both model group and control group were given the same amount of normal saline by gavage from the day of modeling,and the rats in baicalin group were intragastrically given 100 mg/kg baicalin once a day for 30 d from the day of modeling.Heart rate(HR),left ventricular end diastolic pressure(LVEDP)and left ventricular systolic pressure(LVSP),myocardial tissue morphology,apoptosis rate,protease activated receptor 2(PAR-2),B lymphoma-2 gene(Bcl-2)and Bcl-2 related X(Bax)protein expression levels,T lymphocyte subgroup(CD3^(+),CD4^(+),CD8^(+),CD4^(+)/CD8^(+))levels were compared.Results Compared with sham group,the levels of LVSP and Bcl-2 in model group were decreased(P<0.05),while LVEDP,the levels of Bax and PAR-2 proteins were increased(P<0.05).Compared with model group,the heart function,LVSP and Bcl-2 levels were increased in baicalin group(P<0.05),while the levels of LVEDP,Bax and PAR-2 were decreased(P<0.05).Compared with sham group,the levels of CD3^(+),CD4^(+),and CD8^(+)were significantly increased in model group(P<0.05),while CD4^(+)/CD8^(+)was significantly decreased(P<0.05).Compared with model group,the levels of CD3^(+),CD8^(+)were decreased(P<0.05),while CD4^(+)/CD8^(+)was increased in baicalin group(P<0.05),and CD4 showed no significant change(P>0.05).Compared with sham group,the rat cardiomyocyte apoptosis rate was increased in model group(P<0.05).Compared with model group,the rat cardiomyocyte apoptosis rate in baicalin group was significantly reduced(P<0.05).Conclusion Baicalin can improve the cardiac function of rats with myocardial infarction and the immune function of myocardial tissue,and reduce the apoptosis rate of myocardial cells,which may be related to the up-regulation of Bcl-2 and PAR-2 proteins and the down-regulation of Bax protein expression.
