基于数据挖掘和网络药理学研究原发性骨肿瘤的用药规律及作用机制 被引量:2

Bioinformatical and network pharmacological analysis of traditional Chinese medicine actions on primary bone tumors and underlying mechanisms

摘要 目的挖掘中医药治疗骨肿瘤的用药规律,并基于网络药理学探讨高频药物治疗骨肿瘤的药理机制。方法检索中国知网、万方数据知识服务平台、维普网中的文献,使用Excel 2016、SPSS Modeler 18.0、SPSS Statistics 24.0软件,将纳入文献的中药进行聚类分析、关联规则分析,从DisGENT数据库及TTD数据库提取骨肿瘤的主要靶点,TCMSP数据库中提取高频药物的主要成分,通过ChemMapper数据库提取主要成分的预测靶点,使用Cytoscape 3.2.1软件构建成分-靶点网络,进一步分析主要通路及潜在作用机制。结果最高频次用药为黄芪,使用最多的药物类别为补虚药,药物四气五味主要以甘、温为主,归经中以肝经最多,关联规则强度最高的组合有六对,聚类分析可归为补肾填精、化痰利湿散结、补益气血三类。桦木酸为黄芪发挥作用的主要成分,GSK-3β为核心靶点,主要作用于PI3K/Akt信号通路。结论骨肿瘤的治疗多以黄芪为主药,可随证配伍补肾填精、活血化瘀、益气养血等药物,可能通过PI3K/Akt信号通路发挥作用。 Objective To assess traditional Chinese medicine(TCM)actions on primary bone tumor using bioinformatics and network pharmacology approaches and then to explore the underlying mechanisms.Methods Literature was searched in the CNKI,Wanfang,and Weipu Network databases,and the data were retrieved and analyzed using the Excel 2016,SPSS Modeler 18.0,SPSS Statistics 24.0 software for clustering and association rules of different TCMs.The main targets of bone tumors were identified from the DisGENT and TTD databases,while the main components of high-frequency drugs were identified from the TCMSP database and then the main targets of their active ingredients were identified from the ChemMapper database.The composition-target network was constructed using Cytoscape 3.2.1 software for analysis of the major pathways and underlying mechanisms.Results The most frequent TCM used to treat primary bone tumor was Radix Astragali,while the most commonly used decoctions were nourishing-deficiency herbs with the four qi and five flavors that were mainly"sweet"and"warm".The major TCM meridian tropism was the liver meridian.Overall,there were six core TCM decoctions in the treatment of primary bone tumor through the association rules,while the cluster analysis was able to classify them into three categories,i.e.,tonifying kidney and replenishing essence,resolving phlegm and removing dampness,and replenishing qi and blood.Furthermore,Betulinic acid was the main functional component in Radix Astragali and GSK-3βwas the main target of the drug,while GSK-3βis the major player in the PI3K/Akt pathway.Conclusion Radix Astragali is the main TCM in the treatment of primary bone tumors and frequently combines with other TCM herbs to tonify the kidney and replenish the essence,activate the blood circulation,dissipate blood stasis,and replenish the qi and blood,all of which may regulate the activity of the PI3K/Akt signaling.
