摘要
目的观察中药清肝利水方对慢性高眼压大鼠眼压的影响。方法将48只SD大鼠随机分为空白组、模型组、中药低浓度组(LCG)、中药中浓度组(MCG)、中药高浓度组(HCG)、阳性对照组(PG),每组8只,以左眼为实验眼。采用眼前房注射超顺磁性氧化铁磁珠(SIO)建立慢性高眼压青光眼模型。空白组、模型组和阳性对照组饮用水灌胃,中药各浓度组分别以不同浓度清肝利水方水溶液灌胃,每日1次;阳性对照组术眼予盐酸左布诺洛尔滴眼液滴眼,其余各组大鼠术眼予0.9%氯化钠溶液滴眼,每日1次。共给药14 d。于造模前及造模后测量左眼眼压。结果造模后,模型组眼压均高于空白组(t_(1d)=22.259,t_(3d)=19.065,t_(7d)=10.545,t_(14d)=11.204,均P=0.000)。造模后7 d,中药中浓度组眼压低于中药低、高浓度组,差异均有统计学意义(t_(LCG)=58.106,t_(HCG)=30.370,均P=0.000)。造模后14 d,各治疗组眼压均低于模型组(t_(LCG)=20.981,t_(MCG)=14.570,t_(HCG)=18.737,t_(PG)=11.539,均P=0.000);与阳性对照组相比,中药各浓度组眼压均升高,差异均有统计学意义(t_(LCG)=6.671,t_(MCG)=6.570,t_(HCG)=5.279,均P=0.000);中药各浓度组间眼压差异无统计学意义(P>0.05)。结论清肝利水方能够降低SIO造模的慢性高眼压青光眼大鼠的眼压。
OBJECTIVE To observe the effect of Qinggan Lishui formula on intraocular pressure of rats with chronic ocular hypertension.METHODS Forty-eight SD rats were randomly divided into the blank group,the model group,Chinese medicine groups including the low concentration group(LCG),the medium concentration group(MCG),the high concentration group(HCG),and the positive control group(PG)with eight rats in each group,and the left eye was set as the experimental eye.The anterior chamber of the eye was injected with superparamagnetic iron oxide(SIO)to establish the chronic high intraocular pressure glaucoma model.The control group,the model group,and the positive control group were irrigated with drinking water,and each Chinese medicine concentration group was given different concentrations of Qinggan Lishui decoction once a day;the positive control group received levobunolol hydrochloride eye drops for the operation eye,rats of the other groups were given 0.9%sodium chloride solution eye drops once a day.Intervention duration was 14 days.The intraocular pressure of left eye was measured before and after modeling.RESULTS After modeling,the intraocular pressure of the model group was higher than that of the blank group(t_(1d)=22.259,t_(3d)=19.065,t_(7d)=10.545,t_(14d)=11.204,all P=0.000).Seven days after modeling,the intraocular pressure of the medium concentration group was lower than that of the low and high concentration groups,and the difference was statistically significant(t_(LCG)=58.106,t_(HCG)=30.370,all P=0.000).Fourteen days after modeling,the intraocular pressure of each treatment group was lower than that of the model group(t_(LCG)=20.981,t_(MCG)=14.570,t_(HCG)=18.737,t_(PG)=11.539,all P=0.000);Compared with the positive control group,the intraocular pressure of each concentration group increased,and the difference was statistically significant(t LCG=6.671,t_(MCG)=6.570,t_(HCG)=5.279,all P=0.000),There was no statistically significant difference in intraocular pressure between the concentration groups(P>0.05).CONCLUSIONS Qinggan Lishui formula can reduce the intraocular pressure of chronic ocular hypertension glaucoma rats modeled by SIO.
