急性肠梗阻肠坏死患儿A2B腺苷受体、紧密连接蛋白、肿瘤坏死因子-α表达及与肠上皮细胞凋亡的相关性分析 被引量:2

Correlations between Expressions of A2BAR,Occludin and TNF-αwith Intestinal Epithelial Cell Apoptosis in Children with Acute Intestinal Obstruction and Necrosis

摘要 目的分析A2B腺苷受体(A2BAR)、紧密连接蛋白、肿瘤坏死因子-α(TNF-α)在急性肠梗阻肠坏死患儿中的表达及与肠上皮细胞凋亡的相关性。方法将2015年4月—2020年2月于本院行手术治疗的65例急性肠梗阻肠坏死患儿作为研究组,同期于我院诊治的30例消化道畸形患儿作为对照组。比较2组A2BAR、紧密连接蛋白及TNF-α表达情况,并采用Pearson相关分析观察急性肠梗阻肠坏死患儿A2BAR、紧密连接蛋白、TNF-α与肠上皮细胞凋亡指数(AI)的关系。结果研究组A2BAR灰度值、紧密连接蛋白灰度值及AI低于对照组(P<0.05,P<0.01)。与对照组术前比较,研究组术前血清TNF-α水平显著升高(P<0.05);术后7 d,研究组血清TNF-α水平较术前显著降低(P<0.05)。A2BAR、紧密连接蛋白灰度值与AI呈正相关(P<0.01),TNF-α与AI呈负相关(P<0.01)。结论A2BAR、紧密连接蛋白及TNF-α在急性肠梗阻肠坏死患儿中异常表达,且与患儿肠上皮细胞大量凋亡关系密切,临床需重视患儿A2BAR、紧密连接蛋白及TNF-α水平监测,以改善其预后。 Objective To analyze expressions of A2B adenosine receptor(A2BAR),tight junction protein and tumor necrosis factor-α(TNF-α)in children with acute intestinal obstruction and necrosis and their correlations with intestinal epithelial cell(IEC)apoptosis.Methods A total of 65 children with acute intestinal obstruction and necrosis who had undergone operations between April 2015 and February 2020 were enrolled as research group,and other 30 children with digestive tract malformations treated at the same period were selected as control group.Expressions of A2BAR,tight junction protein and TNF-αwere compared between two groups.Correlations between expressions of A2BAR,tight junction protein and TNF-αwith IEC apoptosis index(AI)in children with acute intestinal obstruction and necrosis were analyzed by Pearson correlation analysis.Results Values of A2BAR gray value,tight junction protein and AI in research group were significantly lower than those in control group(P<0.05,P<0.01).Serum TNF-αlevel before operation in research group were significantly increased compared with that in control group before operation(P<0.05).After operation for 7 d,serum TNF-αlevel was significantly lower than that before operation in research group(P<0.05).A2BAR and gray value of tight junction protein positively connected with AI(P<0.01),and TNF-αnegatively connected with AI(P<0.01).Conclusion A2BAR,tight junction protein and TNF-αhave abnormal expressions in children with acute intestinal obstruction and necrosis,which closely relate to apoptosis of a large number of IEC.Monitoring A2BAR,tight junction protein and TNF-αlevels should be paid more attention so as to improve its prognosis.
