摘要
目的探讨心安颗粒对急性心肌梗死大鼠心肌组织的保护及心肌纤维化的干预作用。方法采用左冠状动脉结扎的方法,制备急性心肌梗死后心肌重塑模型。将40只成年正常雄性SD大鼠随机数字表法分为假手术组、模型组、卡托普利组、心安颗粒组,各10只。药物干预8周后,采用电镜、免疫组化等方法检测左心室肥厚指数、心肌胶原容积分数(CVF)、Ⅰ型胶原容积分数(ⅠCVF)、基质金属蛋白酶1(MMP-1)/基质金属蛋白酶抑制剂1(TIMP-1)。结果模型组Ⅰ型胶原蛋白上调,CVF明显升高,且胶原蛋白沉积表现为增加,最终形成了心室重构问题,使得心肌纤维化产生。两组在心肌胶原容积分数方面得到显著的下降,可减轻心肌胶原蛋白沉积程度、抑制心室重构,逆转心肌纤维化的作用。结论心安颗粒可通过下调MMP-1、TIMP-1,调节MMP-1/TIMP-1,调控细胞外基质代谢,从而达到抑制心室重构、逆转心肌纤维化的作用。
Objective To investigate the protective effect of Xin'an granules on myocardial tissue and myocardial fibrosis in rats with acute myocardial infarction.Methods The models of myocardial remodeling after acute myocardial infarction were made by the ligation of left coronary artery.Forty normal male adult SD rats were divided into a sham operation group,a model group,a captopril group,and a Xin'an granule group by the random number table method,with 10 in each group.After the drug intervention for 8 weeks,electron microscope,immunohistochemical method,etc.were used to detect their left ventricular hypertrophy,myocardial collagen volume fraction(CVF),typeⅠcollagen volume fraction(ⅠCVF),and MMP-1/TIMP-1.Results TypeⅠcollagen was upregulated in the model group;the CVF was significantly increased,and the collagen deposition was increased,resulting in ventricular remodeling and myocardial fibrosis.The CVF decreased significantly in both groups,which could reduce the myocardial collagen deposition,inhibit ventricular remodeling,and reverse the myocardial fibrosis.Conclusion Xin'an granules can inhibit ventricular remodeling and reverse myocardial fibrosis by down-regulating MMP-1 and TIMP-1 and regulating MMP-1/TIMP-1 and extracellular matrix metabolism.
