小鼠宫颈癌组织放化疗前后NDRG1蛋白表达差异及预后意义 被引量:2

摘要 目的:探讨小鼠宫颈癌组织中N-myc下游调节基因1(N-myc downstream regulated gene 1,NDRG1)蛋白在放化疗前后的表达差异及预后意义。方法:建立U14荷瘤小鼠模型,根据成瘤后处理方式不同,将40只小鼠随机分为四组:对照组、化疗组、放疗组和放化疗组。测量各组小鼠肿瘤瘤重并计算抑瘤率,采用蛋白质印迹法(Western blot)检测不同处理组小鼠宫颈癌组织中NDRG1蛋白的表达水平。Spearman法对移植瘤瘤重与NDRG1蛋白相对灰度进行相关分析。结果:与对照组相比,处理组移植瘤瘤重较低,差异有统计学意义(P<0.05)。其中放化疗组平均瘤重最低[(0.89±0.47)g]、抑瘤率最高(75.68%)。化疗组、放疗组和放化疗组NDRG1蛋白相对灰度值高于对照组,差异有统计学意义(P<0.05)。NDRG1蛋白相对灰度与移植瘤瘤重呈负相关(r=-0.376,P=0.017)。结论:放疗、化疗及放化疗可引起小鼠宫颈癌组织NDRG1表达量降低(蛋白相对灰度增加),NDRG1表达量改变可以一定程度反映宫颈癌的治疗效果。 Objective:To investigate the difference of N-myc downstream regulated gene 1(NDRG1)expression in mice cervical cancer xenograft tissues before and after radiotherapy and chemotherapy and its prognostic significance.Methods:The mice subcutaneous xenograft model of cervical cancer U14 cells was established and 40 mice were randomly divided into four groups,including control group,chemotherapy group,radiotherapy group and chemoradiotherapy group according to the different ways of treatment after tumorigenesis.The tumor weight and tumor inhibition rate of mice in each group were measured.The expression level of NDRG1 in the cancer tissues of mice was detected by Western blot.The correlation between the tumor weight and the relative gray of NDRG1 protein was analyzed by Spearman's method.Results:Compared with the control group,the average tumor weight of the experimental groups was lower(P<0.05).The chemoradiotherapy group had the lowest average tumor weight[(0.89±0.47)g]and the highest tumor inhibition rate(75.68%)among the four groups.Compared with the control group,the relative gray value of NDRG1 protein in chemotherapy group,radiotherapy group and chemoradiotherapy group was higher,and the difference was statistically significant(P<0.05).The relative gray value of NDRG1 protein was inversely associated with the tumor weight(r=-0.376,P=0.017).Conclusion:Radiotherapy,chemotherapy and chemoradiotherapy can reduce the expression of NDRG1 in mice cervical cancer tissues(the relative gray level of protein was increased),and it can partly reflect the therapeutic effect of cervical cancer.
