摘要
目的通过观察针刺对膀胱过度活动症(OAB)大鼠的治疗效果,探讨针刺治疗对大鼠膀胱功能及膀胱黏膜组织配体门控性非选择性阳离子通道受体(P2X)3蛋白表达的影响。方法将30只大鼠分为对照组、模型组、针刺组,每组10只。模型组与针刺组的大鼠采用近端尿道结扎法建立膀胱流出道梗阻诱导的OAB模型,对照组大鼠采用除结扎尿道外同样的手术操作行假伤对照。针刺组大鼠进行针刺治疗,对照组和模型组不做任何干预治疗。测量收集各组实验大鼠的尿动力学数据,采用荧光定量PCR(RT-PCR)检测膀胱组织P2X3 mRNA相对表达量,用Western印迹法检测膀胱组织P2X3蛋白的相对表达量。结果与模型组相比较,对照组、针刺组的膀胱基压和收缩后膀胱内压均降低(均P<0.05),最大逼尿肌收缩压、膀胱收缩时间和排尿间隔时间均升高(均P<0.05)。与模型组相比较,对照组和针刺组的P2X3蛋白相对表达量均降低(均P<0.05),且与对照组相比较,针刺组的P2X3蛋白相对表达量降低(P<0.05)。膀胱基压和收缩后膀胱内压与P2X3蛋白相对表达量呈正相关(P<0.05),最大逼尿肌收缩压、膀胱收缩时间和排尿间隔时间与P2X3蛋白相对表达量呈负相关(P<0.05)。结论针刺治疗对有效治疗OAB,改善膀胱功能,可能与有效降低P2X3蛋白的表达有关。
Objective By observing the therapeutic effect of acupuncture on overactive bladder(OAB)rats,the effect of acupuncture on the bladder function and the expression of P2X3 protein in bladder mucosa tissue of rats was explored.Methods Thirty rats were divided into control group,model group and acupuncture group,with 10 rats in each group.The rats in the model group and the acupuncture group were established with the proximal urethral ligation method to establish OAB model for bladder outflow tract obstruction induction.The rats in the acupuncture group were treated with acupuncture,and the control group and model group were not treated with any intervention.The urodynamic data of experimental rats in each group were measured,the relative expression of P2X3 mRNA in bladder tissue was detected by fluorescence quantitative PCR,and the relative expression of P2X3 protein in bladder tissue was detected by Western blot.Results The bladder basal pressure,maximum detrusor systolic pressure,bladder systolic time,bladder internal pressure after systole,and micturition interval time were statistically significant among the three groups of experimental rats(all P<0.05).Compared with the model group,the bladder base pressure and the bladder internal pressure after systole in the control group and acupuncture group decreased(both P<0.05),and the maximum detrusor systolic pressure,bladder systolic time,and voiding interval time all increased(all P<0.05).The relative expression of P2X3 mRNA in bladder mucosa tissue of three groups of experimental rats was not statistically significant(P>0.05),and the expression level of P2X3 protein in bladder mucosa tissue of three groups of experimental rats was statistically significant(P<0.05).Compared with the model group,the relative expression of P2X3 protein in the control group and the acupuncture group decreased(both P<0.05),and compared with the control group,the relative expression of P2X3 protein in the acupuncture group decreased(P<0.05).The bladder base pressure and bladder internal pressure were positively correlated with the relative expression of P2X3 protein(P<0.05),and the maximum detrusor systolic pressure,bladder contraction time and urination interval time were negatively correlated with the relative expression of P2X3 protein(P<0.05).Conclusions Acupuncture treatment could effectively treat OAB and improve bladder function,which might be related to the effective reduction of P2X3 protein expression.
